目的

观察含海藻与不同基原甘草反药组合的海藻玉壶汤(HYD)对甲状腺肿大模型大鼠脂质代谢及肝脏的影响。
方法

将96只大鼠用随机数字表法分为空白(Control)组、模型(Model)组、阳性药(Euthyrox)组、HYD-乌拉尔甘草(HYD-U)组、HYD-光果甘草(HYD-G)组、HYD-胀果甘草(HYD-I)组6组,各16只。Control组予相应体积去离子水(1 mL/100 g)灌胃;其余各组给予丙硫氧嘧啶溶液1 mL/100 g灌胃,1次/d,连续灌胃14 d复制甲状腺肿大模型。Control组、Model组予去离子水灌胃,Euthyrox组给予Euthyrox溶液2 μg/100 g灌胃,HYD-U组、HYD-G组、HYD-I组分别给予中药汤剂1.206 g/100 g灌胃,共干预28 d。对比各组血脂水平、病理结构变化及实时定量逆转录聚合酶链反应(RT-qPCR)检测的肝脏相关基因表达水平。

Objective

To observe the effects of
Haizao Yuhu Decoction
(HYD) containing anti-drug combinations of
Sargassum
with different basidiogenetic
Glycyrrhizae Radix et Rhizoma
(GR) on lipid metabolism and liver function in goiter model rats.
Methods

Ninety-six rats were randomly assigned using a random number table to six groups: Control, Model, positive drug (Euthyrox), HYD-Glycyrrhiza uralensis Fisch. (HYD-U), HYD-G. glabra L. (HYD-G), and HYD-G. inflata Bat. (HYD-I), with 16 rats per group. The Control group received deionized water (1 mL/100 g) via gastric gavage. The other groups were given propylthiouracil (PTU, 1 mL/100 g) once daily for 14 days to induce goiter. After modeling, the Control and Model groups continued to receive deionized water, the Euthyrox group received Euthyrox solution (2 μg/100 g), and the HYD-U, HYD-G, and HYD-I groups received the respective herbal decoctions (1.206 g/100 g) via gastric gavage for 28 days. Blood lipid levels, liver histopathology, and hepatic gene expression (detected by RT-qPCR) were compared among groups.

Compared with the Control group, the Model group showed increased total cholesterol (TC), triglycerides (TG), apolipoprotein B (ApoB), very low-density lipoprotein (VLDL), and low-density lipoprotein (LDL), and decreased apolipoprotein A1 (ApoA1) and high-density lipoprotein (HDL) levels (P < 0.05). Compared with the Model group, the Euthyrox group showed lower TC and LDL, and higher HDL (P < 0.05); the HYD-U group showed lower TG and higher ApoA1, ApoB, HDL, and VLDL (P < 0.05); the HYD-G group showed higher ApoA1, ApoB, and HDL (P < 0.05); the HYD-I group showed lower TG, and higher TC, ApoA1, ApoB, HDL, VLDL, and LDL (P < 0.05). Compared with the Euthyrox group, HYD-U showed higher ApoA1, ApoB, VLDL, and LDL (P < 0.05); HYD-G showed higher TC, ApoA1, ApoB, HDL, and LDL (P < 0.05); HYD-I showed higher TC, ApoA1, ApoB, HDL, VLDL, and LDL (P < 0.05). Compared with HYD-U, HYD-G had higher TG, TC, and LDL but lower ApoB and VLDL (P < 0.05), while HYD-I had higher TC and LDL (P < 0.05). Compared with HYD-G, HYD-I showed lower TG ,and higher ApoB (P < 0.05). Histologically, hepatocytes in all treatment groups were arranged uniformly, hepatic lobule structures were clear, and central veins were centered. Inflammatory infiltration was minimal, and local hepatocyte edema or vacuolar lipid accumulation was reduced compared with the Model group. Compared with the Control group, the Model group showed decreased hepatic mRNA expression of stearoyl-CoA desaturase 1 (SCD1), sterol regulatory element-binding protein-1c (SREBP-1c), and peroxisome proliferator-activated receptor alpha (PPARα) (P < 0.05). Compared with the Model group, Euthyrox increased AMPK and PPARα expression (P < 0.05), while HYD-U, HYD-G, and HYD-I increased AMPK, SREBP-1c, and PPARα expression (P < 0.05). Compared with Euthyrox, HYD-U increased SREBP-1c (P < 0.05); HYD-G increased AMPK and SREBP-1c (P < 0.05); HYD-I increased SREBP-1c (P < 0.05). Compared with HYD-U, HYD-G increased AMPK and SREBP-1c (P < 0.05), and compared with HYD-G, HYD-I increased SREBP-1c (P < 0.05).
Conclusion

HYD containing anti-drug combinations of
Sargassum
with different basidiogenetic GR can modulate blood lipid levels, improve hepatic lipid metabolism, and ameliorate liver histopathological changes in goiter model rats.
