Effect and mechanism of Xuesaitong Capsule on angiogenesis in brain tissue of rats with single side carotid artery stenosis
Objective To explore the effect and mechanism of Xuesaitong Capsule on angiogenesis in brain tissue of rats with single side carotid artery stenosis. Methods SD rats were randomly divided into 3 groups: sham group, Model group, Xuesaitong group. Model group and Xuesaitong group were fed with high fat diet for modeling carotid artery stenosis,and then immediately given Xuesaitong Capsule solution( 3. 15 mg/ml),and sham group and model group were given with 3 ml distilled water. All groups were treated once every 24 h for 42 days. Then the brain tissue were obtained by head cutting,HE staining was used to observe the pathological changes of brain tissue,Immunofluorescence to measure MVD,rt-PCR to investigate the mRNA expression of VEGF,and Western blot to detect the protein expression of VEGFR2. Results Compared with the Model group,the morphological structure of neurons of Xuesaitong group was clear,cytoplasmic of nerve cells was light staining,the cytoplasm of fewer neurons was concentrated,and the level of membrane shrinkage was also lower. Compared with Sham group and Model group,the MVD and the expression of VEGF mRNA and VEGFR2 in Xuesaitong group were all increased( P < 0. 05,P < 0. 05,P < 0. 01). Conclusion As a representative drug for activating blood and dissolving stasis,Xuesaitong Capsule can promote angiogenesis in brain tissue of rats with single side carotid artery stenosis,and the mechanism may be related to the activation of VEGF pathway.
