摘要

目的观察甲亢宁胶囊（以下简称甲亢宁）对FRTL-5细胞周期的影响,探讨甲亢宁治疗Graves病的作用机制。方法将FRTL-5细胞分为基线组、模型组、甲亢宁组、甲巯咪唑组,采用MTT法检测甲亢宁不同浓度及作用时间对FRTL-5细胞的影响,分析甲亢宁的量效和时效关系,确定甲亢宁的浓度及作用时间;采用流式细胞技术检测甲亢宁对FRTL-5细胞周期的影响;采用Western Blot检测甲亢宁干预后FRTL-5细胞cyclin D1蛋白的表达水平。结果甲亢宁组处于G0G1期的FRTL-5细胞多于模型组（P <0. 01）,处于S期的FRTL-5细胞少于模型组（P <0. 05）,甲巯咪唑组处于各细胞周期的FRTL-5细胞与模型组间差异无统计学意义（P> 0. 05）。甲亢宁组FRTL-5细胞的cyclin D1蛋白表达水平低于模型组（P <0. 05）,甲巯咪唑组与模型组差异无统计学意义（P> 0. 05）。结论甲亢宁能下调FRTL-5细胞cyclin D1的蛋白表达,阻滞FRTL-5细胞从Gl期进入S期,从而抑制细胞增殖。
