Objective To evaluate the mechanism of anti-hematological tumor of Marsdenia tenacissima extract（ MTE）,and provide evidence for the treatment of hematological tumor with Marsdenia tenacissima and its preparations. Methods THP-1,MV4-11 and HL-60 were target cells,and Marsdenia tenacissima extract was intervention drugs,and through cell culture in vitro,cell proliferation were determined by MTS assay. Flow cytometry was used detect the cell apoptosis and western blot to analyze the related protein expressions. Results MTE could inhibit cell growth of THP-1,HL-60 and MV4-11,especially THP-1,HL-60 cell lines,IC50 were3. 5 mg/m L,3. 02 mg/m L respectively. It could also induce apoptosis in THP-1,HL-60 cell lines. After treatment of 48 h,MTE induced cleavage of PARP in HL-60 and THP-1; caspase-8 in THP-1,and down-regulated of p-Akt in HL-60. Conclusion MTE has a significant anticancer activity in AML cell lines,which could suppress the growth and induce apoptosis in AML cells by activating the expression of caspase 8 and PARP,down-regulating p-Akt.
