Effects of Huangqi TuJian Mixture on the proliferation of glomerular mesangial cells in rats under high-glucose condition and its possible mechanism
摘要
目的探讨黄芪菟箭合剂对高糖状态下大鼠肾小球系膜细胞增殖活性的影响及其可能的机制。方法将雄性SD大鼠随机分为2组,分别以黄芪菟箭合剂及等体积蒸馏水灌胃,以制备含药血清和正常大鼠血清。将大鼠肾小球系膜(HBZY-1)细胞随机分为4组:5 mmol/L葡萄糖+正常血清组(con组),25 mmol/L甘露醇+正常血清组(D-mannitol组),30 mmol/L葡萄糖+正常血清组(HG组),30 mmol/L葡萄糖+含药血清组(HG+HQTJM组)。分别干预24、48、72 h后,应用CCK8法检测各组HBZY-1细胞的增殖活性,Western blot法检测各组HBZY-1细胞内胶原蛋白Ⅳ(CollagenⅣ,COLIV)和纤维连接蛋白(fibronectin,FN)的蛋白表达变化,real time PCR法检测各组HBZY-1细胞内肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白介素-1β(interleukin-1,IL-1β)、人第10号染色体缺失的磷酸酶及张力蛋白同源的基因(phosphatase and tensin homologue deleted on chromosome 10,PTEN)和蛋白激酶B(protein kinase B,PKB/AKT)的mRNA表达变化,Elisa法检测细胞培养上清中TNF-α和IL-1β的蛋白含量。结果与con组比较,D-mannitol组HBZY-1细胞的增殖和细胞内各成分的表达水平差异均无统计学意义(P>0.05);HG组干预48h及72h时HBZY-1细胞增殖活性明显增加,细胞内COLIV和FN蛋白表达增加,细胞内炎症因子TNF-α、IL-1β和AKT基因表达明显增加,差异均有统计学意义(P<0.05),PTEN基因表达水平下降,但差异无统计学意义(P>0.05),细胞培养上清中TNF-α、IL-1β蛋白表达明显增加(P<0.05)。与HG组比较,HG+HQTJM组在干预72h后,可显著抑制HBZY-1细胞增殖(P<0.05),降低细胞内COLIV和FN蛋白的表达;细胞内TNF-αmRNA表达量在干预24h后显著降低(P<0.05),IL-1βmRNA表达量在干预48 h及72 h后均显著降低(P<0.05);在3个干预时间点,HBZY-1细胞内PTEN的基因表达均明显增加,AKT的基因表达明显降低,细胞培养上清中TNF-α及IL-1β蛋白含量明显增加(P<0.05)。结论黄芪菟箭合剂可明显降低高糖状态下HBZY-1细胞的增殖活性,降低HBZY-1细胞内COLIV、FN蛋白的表达,可能与PTEN的激活及PI3K/AKT通路介导的炎症反应受到抑制有关。
Abstract
Objective To investigate the effects of Huangqi Tujian Mixture on the proliferation of glomerular mesangial cells in rats under high-glucose condition and its possible mechanism.Methods Male SD rats were randomly divided into two groups,which were respectively given Huangqi Tujian Mixture and distilled water of equal volume in order to prepare serum contained drug and normal rat serum.The glomerular mesangial(HBZY-1) cells of rats were randomly divided into four groups: 5 mmol/L glucose+normal serum group(control group), 25 mmol/L mannitol + normal serum group(D-mannitol group), 30 mmol/L glucose + normal serum group group(HG group) and 30 mmol/L glucose + drug-contained serum group(HG+HQTJM group).After 24 hours, 48 hours and 72 hours of intervention, CCK8 method was used to detect the proliferation activity and western blot method was used to detect the protein expression changes of collagen Ⅳ and fibronectin in HBZY-1 cells in each group.Meanwhile, the mRNA expressions of tumor necrosis factor-α,Interleukin-1β,phosphatase and tensin homologue deleted on chromosome 10 and protein kinase B in HBZY-1 cells were detected by real time PCR, the expression levels of TNF-α and IL-1 β in the supernatant were measured by Elisa.Results Compared with the control group, D-mannitol group had no significant effect on the proliferation of HBZY-1 cells and the expression of various molecules had no statistical significance(P>0.05), while in the HG group, the proliferation activity of HBZY-1 cells at 48 h and 72 h of intervention increased significantly, the protein expression of COLIV and FN increased, the gene expression of TNF-α, IL-1β and Akt increased significantly(P<0.05), and the mRNA expression of PTEN decreased with the difference not statistically significant(P>0.05), the expression of TNF-α and IL-1β protein in the supernatant of HG group significantly increased(P<0.05).Compared with the HG group, the HG+HQTJM group could significantly inhibit the proliferation of HBZY-1 cells(P<0.05)and the expression of COLIV and fibronectin in mesangial cells after 72 hours’ intervention;the mRNA expression of TNF-α in drug contained serum group was significantly lower than that in HG group after 24 hours’ intervention(P<0.05), and the mRNA expression of IL-1β was obviously lower than that of HG group after 48 hours’ and 72 hours’ intervention(P<0.05).In HBZY-1 cells, the PTEN gene expression significantly increased, Akt gene expression decreased, and TNF-α and IL-1β protein expression in cell culture supernatant increased at three time points of intervention(P<0.05).Conclusion Huangqi Tujian Mixture can significantly reduce the proliferation activity of HBZY-1 cells and down-regulate the protein expression of COLIV, FN in mesangial cells under high-glucose condition, which may be related to the activation of PTEN and the inhibition of inflammation mediated by PI3 K/AKT pathway.
