Effects of acupuncture on Ghrelin in brain tissue and serum,serum IL-1β and IL-4 of model rats with stress-induced ulcer
Objective To observe the effects of acupuncture on the points Guānyuán(关元CV4) and bilateral Xiàjùxū(下巨虚ST39) on the contents of Ghrelin in the brain tissue and serum,serum IL-1β and IL-4 of model rats with stress-induced ulcer(SU),and reveal its mechanism of acupuncture for improving stress-induced ulcer from the angle of endocrine system and inflammatory reaction.Methods A total of 28 SD rats were randomly divided into blank control,model,acupuncture and medication groups,with 7 rats in each group.Except the blank control group,the other three groups were established SU model by water immersion restraint stress.The blank group and model group were bound for 20 minutes every day for 5 days; the acupuncture group got acupuncture on Guānyuán(关元CV4) and bilateral Xiàjùxū(下巨虚ST39) for 20 min with the needles manipulated for 30 s every 5 min,once a day for 5 days.In the medication group,omeprazole enteric-coated tablet solution(0.2 mg/kg) was used,and 2 mL was given by gavage every day for 5 consecutive days.The gastric mucosal damage index(ulcer index,UI) was measured according to Guth’s Methods.Histopathological changes of the gastric mucosa were determined by HE staining.The content of Ghrelin in the serum and brain tissue,IL-1β and IL-4 in the serum were assayed by using ELISA.Results Compared with the blank control group,the gastric mucosal damage index of model group was increased(P<0.01);HE staining showed pathological changes in the gastric mucous; the content of Ghrelin in the serum and brain tissue was decreased(P<0.05); the content of IL-1β in the serum were significantly increased(P<0.01); the content of IL-4 in the serum were significantly decreased(P<0.05) in the model group.Compared with the model group,the gastric mucosal damage index of model group was decreased(P<0.05,P<0.01); HE staining showed the pathological changes in the gastric mucous was improved; the content of Ghrelin in the serum and brain tissue was increased(P<0.05,P<0.01); the content of IL-1β in the serum were significantly decreased(P<0.01); the content of IL-4 in the serum were significantly increased(P<0.01) in the acupuncture and medication groups.The gastric mucosal damage index of acupuncture group was lower than that of the medication group(P<0.05); the content of Ghrelin in the serum and brain tissue of acupuncture group was higher than that of the medication group(P<0.05,P<0.01);the content of IL-1β in the serum of the medication group was higher than that of the acupuncture group(P<0.05); the content of IL-4 in the serum of the acupuncture group was higher than that of the medication group(P<0.05).Conclusion Acupuncture on Guānyuán(关元CV4) and bilateral Xiàjùxū(下巨虚ST39) can relieve mucosal injury in SU rat model through up-regulating the levels of Ghrelin in the serum and brain tissue and IL-4 in serum and down-regulating the levels of IL-1βin the serum and the mechanism might be related with restoration of dynamic balance of endocrine system and maintenance of the relative steady state of inflammatory factors.
