摘要

目的通过文献数据挖掘归纳总结现代中医治疗胃癌常用的药物,利用网络药理学研究方法预测常用药物的作用靶点和分子机制。方法采用计算机检索中国知网（CNKI）、万方、维普和Pub med数据库中医药治疗胃癌的数据挖掘类文献,通过频次分析筛选累计频数较高的前10味中药。SymMap数据库中获取这些药物的靶点,同时在CTD和TTD数据库获得胃癌疾病相关靶点,进一步构建"药物-靶点"和PPI网络,并进行网络拓扑分析筛选关键子模块,再对预测靶点进行GO和KEGG富集分析,探索其背后潜在的分子机制。结果本研究共纳入23项研究,通过筛选获得核心药物:白术、茯苓、白花蛇舌草、半夏、甘草、薏苡仁、陈皮、黄芪、太子参、炒麦芽。网络药理学研究预测得到139个药物靶点,主要涉及细胞因子活性、细胞因子受体结合、泛素样蛋白连接酶结合等分子功能,对活性氧的反应、脂多糖的反应、氧化应激反应等生物学过程,以及膜筏、膜微区、膜区等细胞结构,AGE-RAGE通路、乙型肝炎、胰腺癌等信号通路。关键子模块中包含PTEN、ERBB2、ALB、FASLG、NFKBIA等56个靶点。结论现代中医家治疗胃癌注重补益脾胃之气,同时兼以燥湿化痰、健脾消食。清热解毒类中药以白花蛇舌草使用较多,常用药物可能作用于PTEN、ERBB2、ALB、FASLG、NFKBIA等靶点,通过多通路干预胃癌。
