摘要

目的基于网络药理学的方法探讨附子理中丸治疗抗生素相关性腹泻（antibiotic associated diarrhea, AAD）的作用机制。方法通过中药系统药理学数据库（TCMSP）并检索相关文献获得附子理中丸所含中药的主要化学成分,收集其相对应的靶点;抗生素相关性腹泻的疾病靶点来源于GeneCards、DrugBank、Home-OMIM-NCBI数据库。结合Cytoscape 3.7.1构建药物-核心靶点-有效成分网络、靶标蛋白互作PPI网络。利用Bioconductor中的R包对核心靶标进行GO富集和KEGG通路分析。结果共获得附子理中丸活性化学成分117个,药物靶点661个;获得疾病靶点175个;获得核心靶标25个;PPI蛋白作用网络分析,核心蛋白涉及VEGF、EGFR、AKT1、IL-6等;GO富集分析发现主要参与蛋白磷酸酶结合、磷酸酶结合、细胞因子受体结合等生物学过程;KEGG通路富集发现PI3K-Akt信号通路、AGE-RACE信号通路以及HIF-1信号通路富集明显。结论通过网络药理学分析发现,附子理中丸多成分、多靶点调控机体,通过多途径起到治疗AAD的作用,为附子理中丸的实验研究、临床应用提供了理论支持。
