Effect and mechanism of Yinao Jieyu Decoction on nerve regeneration and repair in rats with depression after cerebral hemorrhage
To explore the effect and mechanism of Yinao Jieyu Recipe on nerve regeneration and repair in depressed rats after cerebral hemorrhage.
Methods

All rats were randomly divided into sham group, sham-operating group, model group, Fluoxetine group, and Yinao Jieyu Decoction group with 8 in each group. The model group, fluoxetine group and Yinao Jieyu recipe group was established depression models by injecting collagenase into caudate putamen to induce cerebral hemorrhage combined with chronic unpredictable mild stimulation (CUMS). On the first day of CUMS, drugs were given to the stomach at the same time. The fluoxetine group was given fluoxetine hydrochloride 2.33 mg/(kg·d), the Yinao Jieyu recipe group was given Yinao Jieyu recipe 9.92 g/(kg·d), and the blank group, sham operation group and model group were given distilled water 10 mL/kg, which was given to the stomach for 4 weeks, body weight and sucrose water test were measured before modeling and at the 1st, 2nd and 4th week, respectively, and the OFT open field experiment was conducted at the 4th week. The expression of Nogo-A, NgR and RhoA protein in hippocampus was detected by Western blotting.

Compared with the blank group and sham operation group, the body weight, sucrose water consumption and the times of standing upright in the model group were less (

0.05). Compared with the model group, the above indexes in fluoxetine group and Yinao Jieyu recipe group were higher (

0.05). Compared with the blank group and sham operation group, the expression of Nogo-A, NgR and RhoA protein in hippocampus of model group was higher (

0.05). Compared with the model group, the expressions of Nogo-A, NgR and RhoA in hippocampus of fluoxetine group and Yinao Jieyu recipe group were lower (

0.05). The expression of Nogo-A protein in hippocampus of Yinao Jieyu recipe group was lower than that of fluoxetine group (

0.05), and the expression of NgR protein was higher than that of fluoxetine group (

0.05). There was no significant difference in the expression of RhoA protein between the two groups (

Yinao Jieyu Recipe can promote nerve regeneration and repair in depressed rats after cerebral hemorrhage by down-regulating the expression of axon growth inhibitory factors Nogo-A, NgR and RhoA.
