Inhibition of p38MAPK signal pathway by Guben Zhike Recipe on airway inflammation in COPD mice
To observe the regulatory effect of Guben Zhike Recipe on airway inflammation in mice with chronic obstructive pulmonary disease (COPD).
Methods
2
Twenty mice were divided into a blank control group, a model group, a Guben Zhike Recipe group, and a dexamethasone group (n=5). Except for the normal group, all other mice were used to established COPD animal models by smoking combined with nasal instillation of lipopolysaccharide (LPS). After successful modeling, the traditional Chinese medicine group was given 0.2 mL/d of Guben Zhike Recipe by gavage of 0.9% physiological saline, while the blank control group and model group were given 0 9% physiological saline was administered by gavage, and the dexamethasone group was given dexamethasone injection (1.0mg/kg) intraperitoneal injection for 28 consecutive days, twice a day. During the modeling period, the general situations of the mice were recorded, and the NO content in lung tissue homogenate was detected using enzyme-linked immunosorbent assay. The expression of p38 MAPK, p-p398MAPK, and ATF-2 proteins was measured using Western blot.
Results
2
Compared with the model group, the NO level in the lung tissue homogenate of Guben Zhike Recipe group (for strengthening the foundation and relieving cough)was decreased(
P
<
0.05), the expression of p38MAPK proteins decreased (
P
<
0.01).
Conclusion
2
Guben Zhike Recipe can improve airway inflammation in stable COPD mice, and the mechanism may be related to the inhibition of the p38 MAPK pathway.
