摘要

目的

探讨健脾益气摄血方对免疫性血小板减少症（Immune thrombocytopenia， ITP）小鼠的治疗作用及机制。

方法

将90只SPF级BALB/c小鼠按随机数字表法分为正常对照组、模型组、强的松组、健脾益气摄血组（中剂量组、高剂量组），每组18只。正常对照组经腹腔注射生理盐水100 μL/20 g，其余各组经腹腔注射抗小鼠血小板血清（APS）100 μL/20 g，1次/d，建立ITP小鼠模型。造模第8天开始，正常对照组、模型组给予0.1 mL/10 g生理盐水灌胃，强的松组及健脾益气摄血中、高剂量组均予0.1 mL/10 g相应药物灌胃，1次/d，共给药8 d。Western blotting法检测构成线粒体电子传递链复合物Ⅱ的琥珀酸脱氢酶黄素蛋白亚基A（SDHA）及调控线粒体蛋白质稳态的关键蛋白酶ClpP、LonP1蛋白表达，荧光定量PCR法检测SDHA、ClpP、LonP1 mRNA表达。

结果

模型组脾脏、心脏与脑组织SDHA蛋白表达高于正常对照组（

0.05），强的松组、健脾益气摄血组脾脏SDHA蛋白表达低于模型组（

0.05）；模型组脾脏、心脏和骨骼肌ClpP蛋白表达均高于正常对照组（

0.05），各治疗组脾脏ClpP表达均低于模型组（

0.05）；模型组脾脏组织LonP1表达低于正常对照组（

0.05），强的松组脾脏组织LonP1表达高于模型组（

0.05），而模型组心脏和骨骼肌LonP1表达均高于正常对照组（

0.05），强的松组骨骼肌LonP1表达低于模型组（

0.05）。蛋白检测结果与相应组织的mRNA转录水平检测结果基本相符。

结论

健脾益气摄血方可调节ITP小鼠线粒体功能，从而改善乏力症状。

Abstract

Objective

To explore the effect and mechanism of Jianpi Yiqi Shexue Prescription （JPYQSX） for immune thrombocytopenia （ITP） mice.

Methods

A total of 90 SPF BALB/c mice were divided into normal control group， model group， prednisone group， Jianpi Yiqi Shexue Prescription （JPYQSX） medium and high dose group according to random number table method with 18 in each group. The normal control group was intraperitoneally injected with 100 μL/20 g of physiological saline， and the other groups were intraperitoneally injected with 100 μ l/20 g of APS once a day to establish ITP mouse models. From the 8th day of modeling， the normal control group and model group were given 0.1 mL/10 g saline by gavage， while the prednisone group and the middle and high dose groups of invigorating spleen and benefiting qi were given 0.1 mL/10 g corresponding drugs by gavage once a day for 8 days. The protein expressions of SDHA， ClpP and LonP1 in tissues and organs of mice were detected by Western blotting； the mRNA expressions of SDHA， ClpP and LonP1 detected by real-time quantitative PCR （qPCR）.

Results

The expression of SDHA protein in the spleen， heart and brain tissue of the model group was higher than that of the control group（

0.05）. The expression of SDHA protein in the spleen of the prednisone group and the JPYQSX group was significantly lower than that of the model group（

0.05）； The expression of ClpP protein in the spleen， heart and skeletal muscle of the model group was significantly higher than that of the control group（

0.05）， and the expression of ClpP in the spleen of each treatment group was significantly lower than that of the model group（

0.05）； the expression of LonP1 in the spleen tissue of the model group was significantly lower than that of the control group（

0.05）， the expression of LonP1 in the spleen tissue of the prednisone group was significantly higher than that of the model group（

0.05）， and the expression of LonP1 in the heart and skeletal muscle of the model group was significantly higher than that of the control group（

0.05）， and the LonP1 expression in skeletal muscle of the prednisone group was significantly low

er than that of the model group（

0.05）. The protein detection results were basically consistent with the transcription level detection results of the corresponding tissues.

Conclusion

JPYQSX can regulate mitochondrial function in ITP mice so as to relieve fatigue.
