摘要

目的

探讨舒肝和胃方治疗慢性萎缩性胃炎（CAG）的作用机制。

方法

将40只Wistar大鼠按照随机数字表法选取10只作为对照组，剩余30只大鼠用复合造模法制备CAG模型，造模成功后，将27只模型大鼠按随机数字表法分为模型组及舒肝和胃方低、高剂量组，各9只。造模24周后，舒肝和胃方低、高剂量组分别给予5、10 g/（kg·d）药物灌胃；模型组、对照组给予等体积灭菌饮用水灌胃，1次/d，连续8周。8周后收集大鼠血清和胃黏膜组织。观察并记录大鼠一般情况、胃黏膜组织病理变化，用RT-qPCR检测大鼠胃黏膜中蛋白酪氨酸激酶2（JAK2）、信号转导和转录激活因子3（STAT3）、c-myc mRNA，免疫组织化学法检测大鼠胃黏膜中JAK2、STAT3、c-myc蛋白，酶联免疫吸附试验检测血清IL-4、IL-10、IL-6。

结果

舒肝和胃方能不同程度改善CAG模型大鼠一般情况。造模20周时，模型组大鼠体质量较对照组低（

0.05）；治疗8周后，舒肝和胃方高剂量组大鼠体质量明显高于模型组和舒肝和胃方低剂量组（

0.05）。舒肝和胃方能改善CAG大鼠胃黏膜状态。与对照组比较，模型组大鼠胃黏膜中JAK2、STAT3、c-myc mRNA表达高（

0.05）。舒肝和胃方高剂量组大鼠胃黏膜中JAK2、STAT3、c-myc mRNA表达均低于模型组和舒肝和胃方低剂量组（

0.05）。与对照组比较，模型组大鼠胃黏膜中p-JAK2、p-STAT3、c-myc蛋白表达高（

0.05）。舒肝和胃方高剂量组大鼠胃黏膜中p-JAK2、p-STAT3、c-myc蛋白表达均低于模型组和舒肝和胃方低剂量组（

0.05）。与对照组比较，模型组大鼠血清IL-6水平高，IL-4、IL-10水平低（

0.05）；与模型组比较，舒肝和胃方低、高剂量组大鼠血清IL-4、IL-10水平高，IL-6水平低（

0.05）；舒肝和胃方低、高剂量组大鼠血清IL-4、IL-10、IL-6水平差异无统计学意义（

0.05）。

结论

舒肝和胃方能修复CAG大鼠胃黏膜，其作用机制可能与抑制JAK2、STAT3、c-myc表达及改变炎症因子IL-4、IL-10、IL-6水平相关。
