Mechanisms of Yixuesheng Capsules in preventing and treating myelosuppression after chemotherapy
in preventing and treating chemotherapy-induced myelosuppression.
Methods
2
Sixty BALB/c male mice were randomly divided into normal, model, intervention A, and intervention B groups using the random number table method. Except for the normal group, mice in the other groups were intraperitoneally injected with cyclophosphamide (100 mg/kg) for 3 days to establish a myelosuppression model. Mice in the intervention A group were gavaged with
Yixuesheng Capsules
(0.39 g/kg) in parallel with modeling, once daily for 21 days. After successful modeling, mice in the normal and model groups were gavaged with saline (0.1 mL per 10 g body mass), while those in the intervention B group were gavaged with
Yixuesheng Capsules
(0.39 g/kg), once daily for 21 days. During the intervention, animal behavior and peripheral blood indices were monitored at different time points. At the end of the experiment, bone marrow cell morphology and nucleated cell counts were observed, and levels of hematopoietic regulatory factors were detected by ABC-ELISA.
Results
2
Compared with the normal group, mice in the model group showed poorer mental state, significantly reduced activity, gray and rough fur, whitened limbs, and weight loss (
P
<
0.05). Compared with the model group, mice in the intervention A and B groups showed significant improvement in mental state and fur condition, and their body weight gradually increased after 7 and 14 days of treatment (
P
<
0.05,
P
<
0.01). After 21 days, body weight in all groups returned to normal levels (
P
>
0.05). Compared with the normal group, red blood cells, hemoglobin, platelets, and leukocytes were significantly decreased in the model group (
P
<
0.01). Compared with the model group, erythrocytes, hemoglobin, and platelets were significantly higher in the intervention A and B groups (
P
<
0.05,
P
<
0.01). Compared with the normal group, bone marrow tissue in the model group showed mass
ive necrosis and a significant reduction in nucleated cells (
P
<
0.01). Compared with the model group, bone marrow nucleated cell counts were significantly higher in the intervention A and B groups (
P
<
0.01). Compared with the normal group, levels of erythropoietin (EPO), thrombopoietin (TPO), granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-11 (IL-11), interleukin-3 (IL-3), and interleukin-6 (IL-6) were significantly decreased in the model group (
P
<
0.05,
P
<
0.01). Compared with the model group, levels of EPO, IL-3, IL-6, and IL-11 were significantly increased in the intervention A group (
P
<
0.05,
P
<
0.01). Compared with the model group, levels of TPO, GM-CSF, IL-11, and IL-3 were significantly elevated in the intervention B group (
P
<
0.05,
P
<
0.01).
Conclusion
2
Yixuesheng Capsules
can improve the general condition of mice with chemotherapy-induced myelosuppression, increase peripheral blood cell counts, and elevate bone marrow nucleated cell numbers. The mechanisms may be related to the regulation of hematopoietic regulatory factor levels.
