Establishment and evaluation of ulcerative colitis rat model of spleen and kidney yang deficiency syndrome
To establish a rat model of ulcerative colitis(UC)with spleen and kidney
yang
deficiency syndrome by intragastric administration of high-dose Senna leaves combined with trinitrobenzene sulfonic acid(TNBS)/ethanol enema,and to validate the model from the perspectives of "prescription testing the syndrome" and "prescription-syndrome discrepancy".
Methods

In the first stage,changes in syndrome and disease indices were evaluated in a blank control group(A)and model rats after 6 weeks of Senna leaf gavage combined with TNBS/ethanol enema.In the second stage,the model rats were randomly divided into a model group (B),a prescription-testing-syndrome group(C),and a prescription-syndrome discrepancy group(D),each with 10 rats.Rats in groups B,C,and D received drinking water,
Wenshen Jianpi Formula
,and
Qingre Huashi Tiaoshu Formula
,respectively,by gavage,and changes in syndrome and
disease indices were further evaluated.

In the first stage, compared with group A,model rats showed reduced food intake,body weight,grip strength,weight-bearing swimming time,and anal temperature (
0.05).In the second stage,compared with group A,rats in groups B,C,and D still had decreased food intake,body weight,grip strength,and weight-bearing swimming time(

Intragastric administration of high-dose Senna leaves combined with TNBS/ethanol enema can successfully establish a UC rat model with spleen and kidney
yang
deficiency syndrome.The modeling procedure is relatively simple to implement and demonstrates high safety.
