Clinical efficacy of Shengxuebao Mixture in treatment of mice with gastric cancer anemia caused by cyclophosphamide
on cyclophosphamide-induced anemia in mice with gastric cancer.
Methods
2
Seventy male BALB/c nude mice were randomly divided into a blank control group, gastric cancer model group, gastric cancer anemia model group, erythropoietin (EPO) group, and low-, medium-, and high-dose
Shengxuebao Mixture
groups according to a random number table, with 10 mice in each group. Except for the blank control group, the gastric cancer model was established in all groups. Except for the blank control group and gastric cancer model group, mice in the remaining groups received intraperitoneal injections of cyclophosphamide at 80 mg/(kg·d) for 4 consecutive days to establish the gastric cancer-related anemia model. The blank control group, gastric cancer model group, and gastric cancer anemia model group received purified water (0.2 mL/d) by gavage for 14 consecutive days. The low-, medium-, and high-dose
Shengxuebao Mixture
groups received
Shengxuebao Mixture
at doses of 2.92, 5.85, and 11.7 mL/(kg·d), respectively, by gavage for 14 consecutive days. The EPO group received purified water (0.2 mL/d) by gavage combined with subcutaneous abdominal injection of recombinant human erythropoietin (rhEPO) at 150 IU/(kg·d) for 14 consecutive days. General condition, body weight, and routine blood parameters of the mice were compared among groups.
Results
2
Peripheral blood hemoglobin (HGB), red blood cell (RBC), white blood cell (WBC), and platelet (PLT) counts in the gastric cancer anemia model group and all treatment groups showed a trend of initial decrease followed by gradual recovery. On treatment
day 7, compared with the gastric cancer anemia model group, HGB levels in the gastric cancer model group and EPO group were significantly increased (
P
<
0.05). On treatment day 14, compared with the gastric cancer anemia model group, HGB levels in the EPO group were significantly increased (
P
<
0.05). On treatment day 7, HGB levels in the medium-dose
Shengxuebao Mixture
group and EPO group were significantly higher than those on treatment day 0 in the same groups (
P
<
0.05). On treatment day 14, HGB levels in all treatment groups were significantly increased (
P
<
0.05). On treatment day 7, compared with the gastric cancer anemia model group, no significant differences in RBC counts were observed among the treatment groups (
P
>
0.05). On treatment day 14, RBC counts in the medium- and high-dose
Shengxuebao Mixture
groups were significantly higher than those in the gastric cancer anemia model group (
P
<
0.05). On treatment day 7, WBC counts in the medium- and high-dose
Shengxuebao Mixture
groups were significantly higher than those in the gastric cancer anemia model group (
P
<
0.05). On treatment day 14, WBC counts in the low-dose
Shengxuebao Mixture
group were significantly higher than those in the gastric cancer anemia model group (
P
<
0.05). On treatment day 7, PLT counts in the low- and medium-dose
Shengxuebao Mixture
groups were significantly higher than those in the gastric cancer anemia model group (
P
<
0.05). On treatment day 14, PLT counts in all treatment groups were significantly higher than those in the gastric cancer anemia model group (
P
<
0.05). Compared with the blank control group, mice in the model groups showed poor mental status, markedly reduced activity, curled posture, grayish-purple skin color, and varying degrees of body weight l
oss. Three days after modeling, reduced appetite and sporadic deaths were observed. Compared with the model groups, the general condition of mice in all treatment groups improved. On treatment days 7 and 14, there were no statistically significant differences in body weight among the treatment groups (
P
>
0.05). Compared with the gastric cancer anemia model group, there were no statistically significant differences in liver, spleen, or kidney indices among the
Shengxuebao Mixture
groups (
P
>
0.05). Compared with the gastric cancer model group, the kidney index in the medium-dose
Shengxuebao Mixture
group was significantly lower (
P
<
0.05).
Conclusion
2
Shengxuebao Mixture
can improve the general condition of mice with gastric cancer-related anemia to a certain extent, increase peripheral blood HGB levels and RBC counts, and also exert beneficial effects on WBC and PLT counts.
