摘要:Objective Network pharmacology can show the characteristics of TCM compound prescriptions with multi-component, multi-target and overall synergistic action and provide a new strategy for the mechanism study of TCM compound prescriptions.Methods In this study, Banxia Xiexin Decoction, a classical prescription to treat chronic atrophic gastritis with definite clinical effect, was observed as a subject.First, the chemical constituents and corresponding drug targets of Banxia Xiexin Decoction were obtained by using the integrated pharmacological database of TCM(TCMIP).Then,chronic atrophic gastritis disease targets were obtained through Human Phenotype Ontology database.The target interaction network was constructed based on protein interaction network(PPI)database, and a candidate target of Banxia Xiexin Decoction for chronic atrophic gastritis was obtained.Finally, GO function analysis and KEGG pathway enrichment analysis of candidate targets were conducted, and the possible biological pathways and processes of Banxia Xiexin Decoction in the treatment of chronic atrophic gastritis were concluded through literature studies.Results Firstly, 420 chemical constituents in the compound were obtained, 1228 predictive targets of drug action were predicted, and the target interaction network of Chinese herbal medicine was constructed and the corresponding GO and KEGG enrichment analysis Results were obtained.Secondly, five disease targets were combined with the predictive targets of Chinese herbal medicine, and the candidate targets of Banxia Xiexin Decoction for chronic atrophic gastritis were predicted.The molecular networks of Chinese herbal medicine-candidate targets contained in Banxia Xiexin Decoction and related molecular networks of Banxia Xiexin Decoction-candidate targets-chronic atrophic gastritis were constructed.The Results showed that Banxia Xiexin Decoction played an important role in regulating the expression of autoimmune factors, reducing inflammation, nerve growth factor, regulating gastrointestinal motility, protecting gastric mucosa, energy metabolism, improving immunity, regulating cholinergic nerve and inhibiting the proliferation of tumor cells.Conclusion The results of network pharmacological analysis combined with the literature of Banxia Xiexin Decoction on chronic atrophic gastritis suggest that the mechanism of action of Banxia Xiexin Decoction may be related to some biological pathways, such as interleukin, cyclooxygenase-2, MAPK cascade and ERK phosphorylation.
摘要:Objective To observe the immediate brain sustained effects of acupuncture at Fēngchí(GB 20)by using resting-state functional magnetic resonance imaging(rsfMRI)technique and explore the brain functional basis of acupuncture at GB20 for regulating visual cortex.Methods 11 healthy subjects randomly received acupuncture or Von Frey filaments(control)at left GB20.The stimulation time was 30 s and the frequency was 60 times/min.fMRI scan was conducted before and immediately after stimulation for 6 min.MATLAB-based DPABI4.3 software was used to analyze rsfMRI data,including fALFF,ReHo,DC for local brain activity changes,as well as the seed point bilateral posterior cingulate cortex(PCC)and the ventral medial prefrontal(VMPFC)for whole brain functional connectivity and observe the immediate brain sustained effect and whole brain functional connectivity after acupuncture at GB20.Results Compared with the tactile stimulation of Von Frey filaments,acupuncture at left GB20 could clearly modulate the right cuneus and right precuneus which involved to the visual function(voxel P value<0.01, cluster P value <0.05).The FC between PCC and inferior parietal lobule(BA7, 39, 40)were enhanced, and the FC of the VMPFC and the right precuneus(BA7)were decreased(voxel P value<0.01, cluster P value<0.05).Conclusion Acupuncture at GB20 could modulate the visual center and adjacent brain regions,as well as the default network,which may be an important brain function basis for the treatment of eye diseases.
摘要:Objective To explore the evaluation value of modified early warning score(MEWS)and acute physiology and chronic health evaluation score(APACHEⅡ)score on prognosis of sepsis patients with different syndrome types in ICU.Methods 187 sepsis patients in ICU of Guang′anmen Hospital of China Academy of Chinese Medical Sciences from January 2016 to December 2017 were scored in TCM syndrome types, MEWS and APACHE Ⅱ.The scores of two different types of syndromes were compared, as well as the mortality of different syndrome types, the two ROC curves drawn, the correlation and the AUC calculated,prognostic evaluation value of scores in two types evaluated;the correlation of scores of types.Results The mortality and two kinds of scores from low to high were the deficiency syndrome,excessive syndrome, syndrome of intermingled deficiency and excess, and the deficiency syndrome and the other two syndromes were statistically different(P<0.05).The two scores of syndrome of intermingled deficiency and excess could effectively predict the death condition, and the correlation was significantly correlated with the Spearman test(P<0.01);the mortality rates before and after the best truncation point of different syndrome type two scores were compared.The difference was statistically significant(P< 0.05).Conclusion Two scores can evaluate the condition and evaluate the prognosis in the evidence and deficiency syndrome.
摘要:Objective To observe the effect of Jianpi Zhidong Decoction(JD)on the expressions of main neurotrophic factors in prefrontal cortex and striatum of Tourette syndrome(TS)rats.Methods 32 male SD rats were randomly divided into blank control group,TS model group,Tiapride group and JD group with 8 rats in each group.TS model rats were established by intraperitoneal injectioning IDPN(250 mg/kg/d)for consecutive 7 days.After modeling, blank control and TS model group were intragastricly administrated with physical saline, and the remaining two groups were administrated with Tiapride(21 mg/kg)and JD(16 mg/kg)respectively for 8 consecutive weeks.The rats were sacrificed after 24 hours of the last dose, and the specimens were taken and stored in liquid nitrogen.The contents of brain-derived neurotrophic factor(BDNF), glial cell-line derived neurotrophic factor(GDNF), vascular endothelial growth factor(VEGF)and transforming growth factor beta1(TGF-β1)were assayed by ELLSA.Results Compared with the blank control group, the contents of four factors in TS model group were decreased significantly(P<0.05);compared with TS model group, the contents of four factors in Tiapride group and JD group were increased significantly(P<0.05);compared with JD group, the four factors in Tiapride group were increased even more obviously(P<0.05).The expressions of above four factors in prefrontal cortex and striatum were basically the same.Conclusion JD could elevate the expressions of neurotrophic factors in prefrontal cortex and striatum caused by IDPN.
摘要:Objective To compare the pharmacological effects of Shenma Yizhi Formula processed with 3 kinds of preparation techniques on learning and memory capability, brain histopathology, blood lipid indexes, oxidative stress indexes, inflammatory factors, cholinergic indexes in anisodamine-induced model rats.Methods 60 SD rats, 30 female and 30 male, were randomly divided into 6 groups with 10 rats in each group.3 Shenma Yizhi Formula groups with 3 different preparation techniques, including No.1 recipe group, No.2 recipe and No.3 recipe, were given the formula with 2.97 g·kg-1·d-1;the positive control group, donepezil hydrochloride tablet, with 0.45 mg·kg-1·d-1;the blank group and the model group, the equal amounts of pure water.After 4 weeks of delivery, the racemic anisodamine hydrochloride(2 mg/kg)was intraperitoneal injected separately to all the rats except for the blank group.The learning and memory ability of all rats was tested by Morris water maze.All rats were anesthetized by chloral hydrate.Serum indexes of lipids, superoxide dismutasee(SOD), glutathione peroxidase(GSH-PX), malondialdehyde(MDA), interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α), nitric oxide(NO), acetylcho1(Ach)and acetylcholinesterase(AchE)were tested, and the pathological morphology of hippocampal CA1 region observed.Results Compared with the model group, TC level of all Shenma Yizhi Formula groups were decreased(P<0.05).SOD activity and NO level of rats in No.1 recipe group were significantly increased(P<0.05).Compared with No.2 recipe group, the passing platform times of rats in No.3 recipe group were significantly increased(P<0.05).Compared with No.2 recipe group and No.3 recipe group, the SOD activity of No.1 recipe group was significantly increased(P<0.05).Compared with No.3 recipe group, the level of TNF-α was decreased in No.1 recipe group and No.2 recipe group, and the level of NO was increased(P<0.05).Conclusion The pharmacological effects of Shenma Yizhi Formula prepared with 3 kinds of techniques are different, and the pharmacological effect indexes of No.1 recipe were better than those of No.2 recipe and No.3 recipe.
