摘要:<正>2018年国际癌症研究机构(International Agency for Research on Cancer,IARC)调查的最新数据显示:乳腺癌的发病率(24.2%)位于全球女性恶性肿瘤的首位,且52.9%的新发病例发生于发展中国家[1]。我国癌症中心2019年的流行病学数据结果显示:乳腺癌的发病率及死亡率分别位于我国女性恶性肿瘤的首位和第5位,且均呈现持续上升态势;5年生存率逐步提高,但仍与发达国家有较大差距。
摘要:Objective To observe the effects of Tangshenning(TSN) on nephrin and synaptopodin of podocyte in diabetic kidney disease(DKD) mice.Methods The DKD mice model was established by feeding 8-week old KK-Ay mice with high-fat diet for 4 weeks and then randomly divided into model group,TSN group and Valsartan group.One more normal control group(NC) was the same age of C57 BL/6 J mice.TSN group were given TSN(raw herbs) with 20 g/(kg·d)to the stomach.Valsartan group was given Valsartan to the stomach with 10 mg/(kg·d). NC group and model group were given equal amount of saline to the stomach.All mice were successively administrated for 12 weeks.After 0,4,8,and 12 weeks,urine was collected and urine protein was measured for 24 hours.After 12 weeks,blood was taken from the apex to measure the levels of serum creatinine and urea nitrogen.Kidney was removed for the ovbservation of pathological changes by HE,PAS,and Masson staining.The protein expression of nephrin and synaptopodin in podocytes was detected by immunohistochemistry,and mRNA expression of nephrin and synaptopodin in podocytes was detected by RT PCR.Results Compared with NC group,24 h proteinuria,serum creatinine and blood urea nitrogen was significantly increased(P<0.05) in model group.Compared with model group,24 h proteinuria,serum creatinine and blood urea nitrogen was significantly decreased(P<0.05) in TSN group and Valsartan group.Compared with NC group,the glomerular mesangial cells of model group were obvious increased,extracellular matrix obviously proliferated.Compared with the model group,the glomerular mesangial cells in TSN group and Valsartan group were obvious decreased,extracellular matrix obviously was alleviated.Compared with NC group,nephrin and synaptopodin of podocyte and mRNA level were significantly decreased in the model group(P<0.05); Compared with the model group,nephrin and synaptopodin of podocyte and mRNA level were significantly increased in TSN group and Valsartan group(P<0.05).Conclusion TSN can reduce proteinuria and ameliorate renal function of DKD mice,the mechanism of which may be related with increasing of nephrin and synaptopodin of podocyte and alleviating of podocyte damage.
摘要:Objective To observe the effects of acupuncture on the points Guānyuán(关元CV4) and bilateral Xiàjùxū(下巨虚ST39) on the contents of Ghrelin in the brain tissue and serum,serum IL-1β and IL-4 of model rats with stress-induced ulcer(SU),and reveal its mechanism of acupuncture for improving stress-induced ulcer from the angle of endocrine system and inflammatory reaction.Methods A total of 28 SD rats were randomly divided into blank control,model,acupuncture and medication groups,with 7 rats in each group.Except the blank control group,the other three groups were established SU model by water immersion restraint stress.The blank group and model group were bound for 20 minutes every day for 5 days; the acupuncture group got acupuncture on Guānyuán(关元CV4) and bilateral Xiàjùxū(下巨虚ST39) for 20 min with the needles manipulated for 30 s every 5 min,once a day for 5 days.In the medication group,omeprazole enteric-coated tablet solution(0.2 mg/kg) was used,and 2 mL was given by gavage every day for 5 consecutive days.The gastric mucosal damage index(ulcer index,UI) was measured according to Guth’s Methods.Histopathological changes of the gastric mucosa were determined by HE staining.The content of Ghrelin in the serum and brain tissue,IL-1β and IL-4 in the serum were assayed by using ELISA.Results Compared with the blank control group,the gastric mucosal damage index of model group was increased(P<0.01);HE staining showed pathological changes in the gastric mucous; the content of Ghrelin in the serum and brain tissue was decreased(P<0.05); the content of IL-1β in the serum were significantly increased(P<0.01); the content of IL-4 in the serum were significantly decreased(P<0.05) in the model group.Compared with the model group,the gastric mucosal damage index of model group was decreased(P<0.05,P<0.01); HE staining showed the pathological changes in the gastric mucous was improved; the content of Ghrelin in the serum and brain tissue was increased(P<0.05,P<0.01); the content of IL-1β in the serum were significantly decreased(P<0.01); the content of IL-4 in the serum were significantly increased(P<0.01) in the acupuncture and medication groups.The gastric mucosal damage index of acupuncture group was lower than that of the medication group(P<0.05); the content of Ghrelin in the serum and brain tissue of acupuncture group was higher than that of the medication group(P<0.05,P<0.01);the content of IL-1β in the serum of the medication group was higher than that of the acupuncture group(P<0.05); the content of IL-4 in the serum of the acupuncture group was higher than that of the medication group(P<0.05).Conclusion Acupuncture on Guānyuán(关元CV4) and bilateral Xiàjùxū(下巨虚ST39) can relieve mucosal injury in SU rat model through up-regulating the levels of Ghrelin in the serum and brain tissue and IL-4 in serum and down-regulating the levels of IL-1βin the serum and the mechanism might be related with restoration of dynamic balance of endocrine system and maintenance of the relative steady state of inflammatory factors.
摘要:Objective To investigate the effect of Xiefei Lishui Granules on spontaneous hypertension rats(SHR) through testing the level of Ang I,Ang II,ACE,renin and aldosterone(ALD)by ELISA to explore the mechanism of anti-hypertensive effects of Xiefei Lishui Granules.Methods 50 SHR of 13-week old were randomly divided into model group,high dose,medium dose,and low dose of Chinese medicine group and captopril group,with 10 rats in each group;10 Wistar-Kyoto(WKY) rats as normal control group.The blood pressure of the tail artery of rats was measured before the experiment and 4 weeks later.The level of Ang I,Ang II,ACE,renin and ALD were tested and analyzed respectively by ELISA after 4 weeks.Results Compared with the model group,the blood pressure of high dose group,medium dose group,low dose group of Chinese herbal medicine and captopril group were decreased after 4 weeks(PP<0.01).The blood pressure of medium dose group and captopril group showed no significant statistical difference after 4 weeks.Compared with the model group,the level of serum renin and Ang I of captopril group,high dose group,medium dose group and low dose group of Chinese herbal medicine were significantly decreased with the difference statistically significant(P<0.01).Compared with the model group,the level of ALD and Ang II of captopril group and medium dose group were much lowered with the difference statistically significant(P<0.01),the level of ALD and Ang II of the high dose group and low dose group of Chinese medicine were lowered with the difference statistically significant(P<0.05).The level of Angiotensin converting enzyme(ACE) showed no difference between the groups.Conclusion Xiefei Lishui Granules can effectively reduce the blood pressure of tail artery of SHR,and the mechanism might be the effect of inhibiting of the renin-angiotensin-aldosterone system.

摘要:目的采用数据挖掘方法分析推拿手法治疗腹泻的选穴特点和规律。方法计算机检索中国知识资源总库(CNKI)、中文科技期刊数据库(VIP)、万方数据知识服务平台(WanfangData)、Web of Science、PubMed收录的推拿穴位治疗腹泻的临床随机对照试验文献并进行筛选,采用频次统计、聚类分析、关联规则方法分析推拿穴位治疗腹泻的选穴规律。结果共纳入文献12篇,推拿穴位治疗腹泻的主要腧穴为关元、气海、天枢、中脘、足三里、大肠俞等,任脉、足阳明胃经和足太阳膀胱经腧穴使用频次最高,所选腧穴主要集中在胸腹部和腰背部,分析得出"关元—气海"和"气海—中脘"为关联度最高,聚类分析将高频选穴聚成5类。结论推拿穴位治疗腹泻以温补元阳、健脾化湿为主,选穴规律符合腹泻的病因病机特点,可为临床治疗提供参考。
