摘要:Objective To observe the improvement of Yisui Granules for anemia and related symptoms in patients with low-and intermediate-risk type Ⅰ myelodysplastic syndromes( MDS).Methods With randomized double-blind,placebo-controlled research method,48 patients with low-and intermediate-risk type Ⅰ MDS were randomly divided into treatment group and control group,with 24 cases in each group.The treatment group was treated with Yisui Granules,1 bag/time,2 times/d,together with the basic western medicine treatment; the control group was treated with the placebo together with the western medicine basic treatment,1 bag/time,2 times/d,continuous treatment for 8 weeks.After the treatment,short-term efficacy indicators,including blood routine,anemia related symptoms and fatigue degree before and after the treatment were analyzed.Results After the treatment,the hemoglobin level of the treatment group was increased in comparison with that before the treatment,and was also significantly higher than that of the control group( all P < 0.05).After the treatment,the total effective rate of the treatment group was higher than that of the control group( P <0.01),and the clinical symptoms scores of the treatment group were lower than those before the treatment and the control group( P <0.001 or P < 0.05).After the treatment,the symptom of dizziness in the treatment group was better than that before the treatment,the symptoms of shortness of breath,fatigue,dizziness,and numbness in the treatment group were better than those in the control group( P < 0.01 or P < 0.05).After the treatment,the total effective rate of fatigue in the treatment group was higher that of the control group and also lower than that before the treatment( all P < 0.01).Conclusion Yisui Granules can increase the hemoglobin level of patients with low-and intermediate-risk type Ⅰ MDS,and improve the patients’ symptoms and fatigue.
摘要:Objective To explore the mechanism of Indirubin and tanshinone Ⅱa in promoting the degradation of mutant PMLA216TRARα by realgar.Methods Different concentrations of realgar were acted on wild type and mutant PMLA216T-RARα to observe the degradation of PML-RARα in two groups of cells to verify that PMLA216T-RARα is resistant to arsenic.The concentration of each component was determined according to the IC50 value measured by CCK8,and different concentrations and different combinations of realgar,indirubin,and tanshinone Ⅱa were used to act on the transfected PMLA216T-RARα cells.Western blotting was used to detect the effects of single use,pairwise combination and triple combination on PML-RARα degradation and autophagy-related factors( pm TOR,LC3 B).The proteasome inhibitor carfilzomib and the autophagy inhibitor bafilomycin A1 were used to detect the degradation of PML-RARα in each group of cells by Western blotting.Results Compared with the control group( without drug intervention),when the concentration of realgar was 0.5 μmol/L,which was acted on PML-RARα wild type and mutant PMLA216T-RARα,the expression of PML-RARα were significantly degra ded( both P < 0.01).Moreover,the comparison between the groups showed that the expression of PML-RARα of mutant PMLA216T-RARα was higher than that of wild type( all P < 0.01).Compared with the control group,when realgar was 4 μmol/L,indirubin was 8 μmol/L,and tanshinone Ⅱa was 8 μmol/L,and were acted on PMLA216T-RARα respetively,they could all degrade PML-RARα( all P < 0.01) and reduce the level of p-mTOR( all P < 0.01),increase the level of LC3 B( P < 0.05 or P < 0.01).The above effect was the strongest when realgar + tanshinone Ⅱa + indirubin was combined( P < 0.01).Compared with the control group,when realgar + tanshinone Ⅱa + indirubin,realgar + tanshinone Ⅱa + indirubin + carfilzomib were used on PMLA216TRARα,PML-RARα were significantly degraded( all P < 0.01).Compared with the control group,carfilzomib alone,bafilomycin A1 alone,realgar + tanshinone Ⅱa + indirubin + bafilomycin A1,when acting on PMLA216T-RARα,all caused PML-RARα to a significant accumulation,and the expression of PML-RARα in the realgar + tanshinone Ⅱa + indirubin + bafilomycin A1 group was higher than that in the carfilzomib alone and bafilomycin A1 group( all P < 0.01).Conclusion The combination of indirubin and tanshinone Ⅱa can enhance the degradation effect of realgar on mutant PMLA216T-RARα through autophagy,and the combination of the three has the strongest autophagy effect,which may be one of the mechanisms of Chinese medicinal formula Realgar-Indigo naturalis in the treatment of acute promyelocytic leukemia.
摘要:Objective To explore the effect of Yizhi Jiedu Decoction on the learning and memory ability,glucose and lipid metabolism and insulin secretion in diabetic rats.Methods Forty-eight SD rats were randomly divided into normal control group,diabetes model group,low-dose,medium-dose,high-dose Yizhi Jiedu Decoction groups,and donepezil hydrochloride groups,each with eight rats.Except for the normal control group,the other five groups were given a high-sugar and high-fat diet combined with streptothricin to induce diabetes.low-dose,medium-dose,high-dose Yizhi Jiedu Decoction groups were given orally by 0.9,1.8,3.6 g/( kg·d) respectively,donepezil hydrochloride group was given donepezil hydrochloride by 0.45 mg/( kg · d).After four weeks of continuous gastric gavage,the rat body weight,fasting insulin( FINS),fasting blood glucose( FBG),serum total cholesterol( TC),low-density lipoprotein cholesterol( LDL-C),high-density lipoprotein cholesterol( HDL-C),and triglycerides( TG) were measured, and the behavioral changes of rats and the pathological changes of hippocampal neurons were observed.Results Compared with the diabetes model group,the weight of the rats in the high-dose Chinese medicine group was controlled( P < 0.05),FBG,TG,and LDL-C were significantly reduced( P < 0.01,P < 0.05); the rats’ escape latency was significant shortened( P < 0.05),and the number of crossing platforms increased( P < 0.05); pathology suggested that the nucleus pyknosis was decreased,the number of degenerated necrotic cells decreased,and the number of Nissl bodies increased.Conclusion Yizhi Jiedu Decoction can significantly improve the glucose and lipid metabolism disorder,promote the secretion of insulin,and improve the cognitive dysfunction of diabetic rats.

摘要:Objective To observe the protective effect of Qihuo Yin on muscle tissue of rats after acute exhaustive exercise and its influence on behavioral changes,and to explore its anti-fatigue mechanism.Methods Eighteen male Sprague-Dawley rats were randomly divided into blank control group,model group and the Qihuo Yin group,with 6 rats in each group.Treadmill exercise was used to establish exhaustive exercise fatigue model of rats.The blank control group was given the same volume of normal saline by gavage once a day as the rats in the model group,and the rats in the Qihuo Yin group were given Qihuo Yin by gavage once a day; the course of treatment was 7 consecutive days for all groups.The blank control group were not given any training.The rats in the model group and the Qihuo Yin group were given treadmill training for 30 minutes( 15-30 m/min) 30 minutes after gavage for 6 consecutive days,and on day7,they were given treadmill training with a speed of 30 m/min.Exhaustive exercise time was recorded for the rats in the model group and the Qihuo Yin,and the serum levels of creatine kinase( CK),lactate dehydrogenase( LDH),lactic acid( LA),urea nitrogen( BUN) were measure.At the same time,the behavioral changes of rats were observed.Results Compared with the blank control group,the levels of serum LDH,LA,CK,BUN in the model group were significantly higher than those in the blank control group( P< 0.01).Compared with the model group,the exhaustive exercise time of rats in Qihuo Yin group was significantly longer than that in the model group( P < 0.01),and the levels of serum LDH,LA,CK,BUN were significantly lower than those in the model group( P <0.05); there was no significant change in the incubation period of the rats rotating rod test animals in the Qihuo Yin group,but the activity latency of the total area of the autonomic activity reaction box was significantly prolonged( P < 0.05).Conclusion Qihuo Yin can significantly prolong the exhaustive exercise time of rats and have an effect on the autonomic activity of rats.It plays an anti-fatigue effect by reducing the production of various metabolites during exercise-induced fatigue in rats.
