摘要:ObjectiveTo examine the improvement mechanism of mitochondrial biosynthesis in HL-1 cardiomyocytes mediated by fast pacing with Qipo Shengmai Granules (QPSM) contained serum.MethodsAn atrial fibrillation cell model with fast pacing was established in vitro by electrical stimulation of HL-1 atrial myocytes. Subsequently, the expression of PGC-1α gene was silenced by transfection of small interfering RNA (siRNA), and were treated with QPSM-contained serum or blank serum. By detecting the number of mitochondria, ATP synthesis ability and the expression of mitochondrial biosynthesis-related proteins, it was observed that when PGC-1α gene was silenced, whether QPSM could still play a role in protecting and promoting the synthesis of cardiomyocyte mitochondria.ResultsThe QPSM-contained serum could increase the number of mitochondria and ATP content, and up-regulate the proteins related to mitochondrial biosynthesis in rapidly pacing HL-1 cardiomyocytes. After transfection with PGC-1α siRNA, the mitochondrial protective effect of QPSM was blocked.ConclusionIt is suggeted that QPSM may improve mitochondrial malfunction of fast paced HL-1 cardiomyocytes through PGC-1/NRF-1/Tfam-mediated mitochondrial biosynthesis.
摘要:ObjectiveTo summarize TCM syndrome characteristics of coronary artery disease (CAD) patients accompanied with persistent atrial fibrillation (PeAF).MethodsA total of 282 CAD patients with PeAF were selected and the symptoms and signs of these patients were collected by four diagnostic methods, then TCM syndromes were performed cluster analysis.ResultsChest tightness, palpitation, insomnia and fatigue were the most common symptoms of CAD accompanied with PeAF, dark red tongue and yellow greasy tongue were common tongue manifestations, and thin wiry pulse and wiry smooth pulse are common pulse conditions. There were four common TCM syndromes, that is Qi and Yin deficiency syndrome complicated with phlegm and blood stasis, deficiency of liver and kidney, Yin deficiency and fire flourishing syndrome, blood stasis and water retention syndrome.ConclusionThe main TCM syndrome of CAD patients with PeAF is Qi and Yin deficiency together with phlegm and blood stasis. To some extent, the research results could reflect the clinical syndrome distribution characteristics of CAD accompanied with PeAF, which is helpful to guide the clinical syndrome differentiation and treatment.
摘要:ObjectiveTo explore the potential biomarkers related to serum bile acids for coronary heart disease combined with paroxysmal atrial fibrillation (CHD-PAF) patients with heart-spleen yang deficiency and stagnant phlegm syndrome.MethodsThe concentration and contents of 43 kinds of serum bile acids (BAs) in CHD-PAF patients with stagnant phlegm syndrome and non-stagnant phlegm syndrome and healthy population were detected by ultra-high performance liquid chromatography-mass spectrometry. Partial least square discriminant analysis (PLS-DA) was used to compare the metabolic profile of serum BAs among three test groups. Correlation analysis was used to analyze the correlation between serum BAs and the clinical phenotype, as well as BAs and occurrence of stagnant phlegm syndrome, and to screen the potential biomarkers related to serum BAs for CHD-PAF patients with stagnant phlegm syndrome.ResultsCompared with CHD-PAF patients with non-stagnant phlegm syndrome, the serum bile acid level was significantly higher than that of patients with non-stagnant phlegm syndrome, and it was correlated with the occurrence of CHD-PAF phlegm syndrome. With the strongest correlations being found for total, secondary and unconjugated BAs in particular.ConclusionElevated serum total BAs, secondary BAs and unconjugated BAs can be used as potential biomarkers for the clinical pattern identification of CHD-PAF patients with heart-spleen yang deficiency and stagnant phlegm syndrome.
摘要:ObjectiveTo observe the clinical efficacy of Jianpi Lishi Tongluo Decoction in the treatment of type 2 diabetes patients with hyperuricemia.MethodsAccording to the random number table method,72 patients with type 2 diabetes patients complicated with hyperuricemia, who visited Outpatients and wards of Department of Nephrology and Endocrinology, Beijing Hospital of Traditional Chinese Medicine, affiliated to the Capital Medical University, were selected and divided into treatment group and control group,36 patients in each group. The two groups of patients were treated with standardized diabetes treatment, the treatment group plus Jianpi Lishi Tongluo Decoction,one pachage per day,once in the morning and once in the evening;the control group was given benzbromarone,50 mg/day,once a day. The curative effects of TCM syndromes and uric acid (UA) were compared between the two groups. Fasting blood glucose (FBG), 2 h PBG, HbA1C, UA, NO, Xanthine oxidase (XOD), hs-CRP, IL-6 and TCM symptom score in treatment group and control group before and after treatment were observed and compared.ResultsThe total effective rate of TCM syndromes in the treatment group was higher than that in the control group, there was no statistically significant difference in UA total effective rate(P>0.05). After treatment,the indexes of FBG, 2 h PBG, HbA1C,UA, XOD, hs-CRP and IL-6 and the TCM syndrome scores in the treatment group were significantly lower than those in the control group (P<0.05),and the content of NO was significantly higher than that in the control group (P<0.05).ConclusionCompared with benbromarone group,Jianpi Lishi Tongluo Decoction is safe and effective in the treatment of type 2 diabetes patients with hyperuricemia,and its therapeutic mechanism may be related to oxidative stress and inflammatory response.
摘要:ObjectiveTo explore the clinical effect and safety of Qinggan Jiangya Capsules combined with Carvedilol Tablets for hypertensive patients with chronic increased heart rate of stagnated heat of liver meridian syndrome.Methods122 patients with grade 1 and 2 hypertension with resting heart rate >80 beats /min of stagnated heat of liver meridian syndrome admitted to the Cardiovascular Department, Guang’anmen Hospital, China Academy of Chinese Medical Sciences from October 2018 to July 2020 were selected and randomly divided into single-drug group and combined-therapy group according to the random number table method. Single-drug group was prescribed Carvedilol 10mg Bid and combined-therapy group was prescribed Carvedilol 10 mg Bid and Qinggan Jiangya Capsules 0.9 g three times a day. Those whose blood pressure didn't reach target control were prescribed Carvedilol 20 mg Bid after two weeks and the whole course was four weeks. The blood pressure, heart rate, TCM syndrome score, curative effect and safety were compared between the two groups before and after treatment.ResultsAfter two and four weeks, not only SBP and DBP, but also rest heart rate in two groups were decreased significantly(P<0.05). After 4 weeks of treatment, SBP in the combined group was lower than that in the single drug group (P<0.05), the blood pressure compliance rate of combined group was higher than that of single drug group (P<0.05). There was no significant difference between the two groups in heart rate compliance rate and the ratio of Carvedilol to 20 mg twice a day (P>0.05).TCM syndrome scores of both groups were significantly decreased after treatment (P<0.05).The TCM syndrome scores of combined-therapy group patients were significantly lower than those of single-drug group patients(P<0.05).The total effective rate of TCM syndrome in combined-therapy group was significantly higher than that in single-drug group and the difference was of statistical significant(P<0.05).There were no adverse reactions in both groups after treatment.ConclusionQinggan Jiangya Capsules combined with Carvedilol can improve the blood pressure standard rate of patients with grade 1 and grade 2 hypertension accompanied with chronic heart rate increase, and improve the clinical symptoms of patients. The curative effect is better than that of Carvedilol alone.

摘要:ObjectiveTo observe the effect of Qingre Jianpi Tiaochang Recipe in mice with DSS-induced colitis and the expression of spleen T lymphocytes, and to explore the mechanism of the recipe in treating DSS-induced colitis.Methods36 SPF C57BL/6 mice were selected and divided into normal group, model group, control group, high, medium, and low doses of Chinese medicine treatment group with 6 in each group. Except the normal group, the other groups of mice were given 2.5% DSS solution to induce ulcerative colitis model. After the molding is finished, the three Chinese medicine treatment groups were intragastrically administered with high, medium, and low doses of Qingre Jianpi Tiaochang Recipe, the control group was intragastrically administered with Salazine sustained-release granule suspension 0.52 g/(kg·d), the normal and model groups were intragastrically administered with an equal volume of physiological saline. All groups got intervention for 7 days. Food intake, body weight, DAI, colon length and histopathology of mice in each group were observed. Serum levels of cytokines TNF-α, IL-10 and C-reactive protein (CRP) were determined by Elisa.The expression of Treg, Th17, CD4+ and CD8+ cells in mouse spleen was detected by flow cytometry.ResultsFood intake and body weight were increased, colonic morphology restored, the pathological injury improved and DAI reduced, the proportion of Treg and CD4+ cells in spleen of mice increased, while the proportion of Th17 and CD8+ cells decreased in the medium and high doses of Chinese medicine groups. Compared with model group, the difference was not statistically significant (P>0.05); compared with the model group, the serum TNF-α and CRP contents were lower (P<0.05) and the IL-10 content was higher (P<0.05) in the medium and high dose groups of Chinese medicine.ConclusionQingre Jianpi Tiaochang Recipe can restore the body weight, increase food intake, decrease DAI score and reduce intestinal mucosal injury in UC mice.The mechanism may be mediated by regulating the expression and balance of spleen T lymphocytes and related cytokines in UC mice, so as to play an immunomodulatory role.
