      摘要:ObjectiveTo explore whether Qiditangshen Formula can inhibit MPC5 endoplasmic reticulum stress (ERS) induced by high glucose through IRE1α/XBP1s pathway.MethodsMPC5 cells were studied and CCK-8 method was used to screen the concentration and time of high glucose model and the best concentration of Qiditangshen Formula intervention. MPC5 cells were divided into control group,model group and Qiditangshen group. They were cultured in complete medium,35 mmol/L high sugar medium, 35 mmol/L high sugar medium+ Qiditangshen, respectively. Western blot was used to detect the protein expression changes of TXNIP, NLRP3 and endoplasmic reticulum stress related pathway, such as p-IRE1α,IRE1α, XBP1s,p-PERK,PERK,ATF6 protein in podocytes.ResultsCompared with the control group, the expression levels of p-IRE1α/IRE1α,p-PERK/PERK,XBP1s,TXNIP,NLRP3 proteins were significantly higher in the model group of MPC5 cells(P<0.01,P<0.05),the expression of ATF6 had an upward trend but the difference had no statistical significance(P>0.05). Compared with the model group, Qiditangshen Formula group could significantly reduce p-IRE1α/IRE1α,XBP1s,TXNIP,NLRP3 proteins expression level of MPC5 cells induced by high glucose(P<0.01,P<0.05), but the differences in expression of p-PERK/PERK and ATF6 protein had no statistical significancece(P>0.05).ConclusionQiditangshen Formula can improve the ERS of MPC5 cells induced by high glucose,and the mechanism is closely related to the inhibition of the overexpression of IRE1α/XBP1s pathway and TXNIP,NLRP3 proteins downstream.

      摘要:ObjectiveTo observe the effect of Tangbikang on the expression of phosphorylated c-Jun N-terminal kinase (p-JNK) and B-cell lymphama-2 (Bcl-2) in sciatic nerve of diabetes rats.Methods10 male SD rats were randomly selected from 70 male SD rats as the normal group, and the other 60 rats were given high-fat diet and low-dose streptozotocin (STZ) to induce type 2 diabetic rat model. After modeling,diabetic rats were randomly divided into 4 groups: model group, α- Alpha lipoic acid (ALA) group[0.0268g/(kg·d)], Tangbikang high dose group[2.5 g/(kg·d)], Tangbikang low dose group [1.25 g/(kg·d)]. After 16 weeks, morphological changes were observed by HE staining. The proteins expression of p-JNK and Bcl-2 were detected by Western blot. The expression of Bcl-2 mRNA was detected by real-time PCR.ResultsAfter 16 weeks of treatment, compared with the model group, the structure of sciatic nerve fibers in ALA group,Tangbikang high group and Tangbikang low dose group were more regular, and the degeneration and breakage of nerve fibers were less, and their morphology were not much changed. The expression of p-JNK protein in ALA group, Tangbikang high and low dose groups were significantly lower than that in model group (P<0.05,P<0.01). The expression of Bcl-2 protein in sciatic nerve of rats in Tangbikang high dose group was higher than that in model group (P<0.05). The expression of Bcl-2 protein in sciatic nerve of rats in ALA group and low dose group were higher than that in model group, but there were no statistical significance (P>0.05). The expression of Bcl-2 mRNA in ALA group, Tangbikang high and low dose groups were significantly higher than that in model group (P<0.01).ConclusionTangbikang can down regulate the expression of p-JNK protein and up regulate the expression of Bcl-2 protein and mRNA in sciatic nerve of DPN rats,which may be one of the targets of Tangbikang in protecting sciatic nerve of DPN rats.

      摘要:ObjectiveTo investigate the mechanism of Tangluoning for alleviating pyrodeath of sciatic nerve cells in KK-Ay diabetic mice by SIRT1.MethodsTen SPF male C57BL/6J mice aged 8 weeks with ordinary feed were selected as blank control group and 30 SPF male KK-Ay mice aged 8 weeks were fed with high fat diet for 4 weeks. Random blood glucose ≥16.7mmol/L was detected as diabetic mouse model was successfully established.The successfully modeled mice were randomly divided into three groups:model group,low-dose Tangluoning group and high-dose Tangluoning group.The low-dose Tangluoning group was given intragastric administration of crude drug 10g/(kg•d),the high-dose Tangluoning group was given intragastric administration of crude drug 20g/(kg•d),the blank control group and the model group were given intragastric administration of equal volume distilled water, free diet during the experiment, continuous administration for 12 weeks.The morphological structure of sciatic nerve cells was observed by transmission electron microscopy,and the expression levels of SIRT1 and scorched-death related molecules (Caspase1, NLRP3, GSDMD) were detected by Western blot.ResultsCompared with the blank control group, the expression level of SIRT1 protein in model group was significantly down-regulated(P<0.05), the protein expression levels of Caspase1, NLRP3 and GSDMD increased (P<0.05);compared with the model group, SIRT1 protein expression in Tangluoning high-dose group was significantly increased (P<0.05),Caspase1, NLRP3 and GSDMD protein expression levels were decreased (P<0.05).ConclusionTangluoning can reduce scorch death of sciatic nerve cells in DPN mice,and part of its mechanism may be realized through up-regulation of SIRT1, and the effect of high-dose Tangluoning is more obvious.

      摘要:ObjectiveTo explore the effect and mechanism of Qingshi Zhiyang Ointment on inflammation model of human immortalized keratinocytes (HaCaT) and epidermal keratinocyte induced by tumor necrosis factor-α (TNF-α)/interferon-γ (IFN-γ).MethodsHaCaT cells and human epidermal keratinocytes were divided into blank group, model group and experimental group respectively. The blank group received no intervention measures. The model group was stimulated with TNF-α(10 ng/mL)+IFN-γ(10 ng/mL) for 6 h to induce inflammation model. On the basis of modeling, the experimental group was cultured with 0.000 1, 0.001, 0.01 and 0.1 g/mL of Qingshi Zhiyang ointment, and divided into four subgroups according to the above concentrations. The effect of Qingshi Zhiyang ointment on cell viability was determined by cell counting kit -8 method. Quantify microRNA-145 (miRNA-145)was detected by real-time reverse transcriptase polymerase chain reaction(real-time RT-PCR) method,Western Blot (WB) method to detect the expression of Sema3A protein,enzyme linked immunosorbent assay (ELISA) method to determine the expression level of interleukin-31 (IL-31), NGF, nerve growth factor 4 (NT-4).ResultsHaCaT cells: Qingshi Zhiyang Ointment with different concentrations has no obvious inhibition on cell proliferation. Compared with the blank group, the levels of miRNA-145 was decreased and Sema3A protein increased, and the levels of IL-31, NGF, and NT-4 increased (P<0.05). Compared with the model group, the expression of miRNA-145 in 0.001 g/mL group, 0.01 g/mL group and 0.1 g/mL group all increased (P<0.05). Sema3A protein expression decreased (P<0.05). The levels of IL-31, NGF and NT-4 in cell culture supernatants of 0.001 g/mL, 0.001 g/ml, 0.01 g/mL and 0.1 g/mL groups decreased gradually, and the differences among the latter three groups were statistically significant (P<0.05). Human epidermal keratinocytes : compared with the blank group, the expression of miRNA-145 in the model group was lowered, the expression of Sema3A protein was increased, and the levels of IL-31, NGF and NT-4 in cell culture supernatant were higher (P<0.05). Compared with the model group, the expression of miRNA-145 in 0.01 g/mL group and 0.1 g/mL group was increased (P<0.05). The expression of Sema3A protein in 0.001 g/mL, 0.01g/mL and 0.1 g/mL groups was decreased (P<0.05). The levels of IL-31, NGF and NT-4 in cell culture supernatants of 0.01 g/mL and 0.1 g/mL groups were decreased (P<0.05), while the levels of IL-31 and NGF in cell culture supernatants of 0.001 g/mL and 0.001g/ml groups were decreased (P<0.05).ConclusionQingshi Zhiyang Ointment can significantly improve the inflammation of HaCaT cells and human epidermal keratinocytes. The mechanism may be through regulating the level of Sema3A/NGF and related molecules to reduce inflammation.

      摘要:ObjectiveTo observe the effects and mechanism of Siwu Humai Decoction on the injury of human umbilical vein endothelial cells (HUVEC) induced by homocysteine (HCY).MethodsAfter conventional culture and passage of HUVEC,MTT assay was used to detect the cell viability and determine the best experimental concentration of the drug. The cells were divided into groups: group A was normal HUVEC, group B to F was added with 125 μmol/L HCY 100 μL to make cell injury model, after 48 hours of culture, group C was added with 31.25 μmol/L NaHS 100 μL, and groups D, E and F were added with 1.6, 2.2 and 2.8 g/L Siwu Humai Decoction 100 μL respectively, and continued to culture.MTT method was used to detect cell viability of each group, flow cytometry to detect apoptosis, transwell method to detect cell migration rate in each group;nitrate reductase method to detect NO level in cell culture medium. Western blot to detect PI3K, p-AKT, CAV-1, eNOS protein relative expression in each group, and BCA measurement of endothelial cell-monocyte adhesion rate.ResultsThe optimal concentrations of HCY and NaHS were 125 μmol/L and 31.25 μmol/L, respectively. The intervention concentration of Siwu Humai Decoction was selected as 1.6, 2.2, and 2.8 g/L. After HCY intervention, the cell viability values were decreased, apoptosis rate increased, migration rate decreased, NO level decreased,eNOS,PI3K,p-AKT protein relative expression decreased, CAV-1 protein relative expression increased, eNOS mRNA relative ct value decreased and adhesion rate with monocytes increased, while Siwu Humai Decoction could reverse the above expressions(P<0.05),and is concentration-dependent.ConclusionSiwu Humai Decoction can protect HUVEC after HCY injury by regulating the PI3K-AKT-eNOS signaling pathway, increasing NO expression, reducing endothelial cell apoptosis, and improving endothelial cell viability and migration, inhibiting monocyte adhesion, thus reducing endothelial cell damage and achieving the purpose of protecting endothelial cells.

      摘要:ObjectiveTo analyze the distribution and characteristics of TCM syndromes in patients with obstructive sleep apnea (OSA).Methods60 patients, who were diagnosed with OSA during hospitalization from January 2020 to January 2022 in the General Department of Beijing Hospital of Traditional Chinese Medicine affiliated to the Capital Medical University, were collected. The gender,age, TCM syndromes, apnea-hypopnea index (AHI),minimum blood oxygen saturation at night (SaO2), and complicated underlying diseases were summarized.ResultsAmong 60 OSA patients, 33 cases (55.00%) were characterized by mutual obstruction of phlegm and stasis, 13 cases (21.67%) were characterized by dampness and heat obstruction in the middle jiao, 8 cases (13.33%) were characterized by lung and spleen qi deficiency, and 6 cases (10.00%) were characterized by lung and kidney Yang deficiency. Mild OSA was more likely to occur in lung and kidney Yang deficiency than other syndromes(P< 0.05). Severe nocturnal hypoxemia was more likely to occur in the phlegm-stasis syndrome group than in the other syndrome groups (P<0.05). 65.00% patients were complicated with hypertension, and the syndrome of mutual obstruction of phlegm and stasis together with hypertension was more than that of other syndromes(P<0.05).ConclusionAmong the OSA patients,the syndrome of mutual obstruction of phlegm and blood stasis and dampness and heat obstruction in the middle jiao are the most common. Most patients with phlegm and blood stasis syndrome suffer from moderate to severe OSA,more severe hypoxemia at night,and higher risk of hypertension. Patients with lung and kidney Yang deficiency syndrome are more likely to develop mild OSA.

      摘要:ObjectiveTo explore the clinical effect of modern five elements health exercise as adjuvant treatment of H-type hypertension.MethodsUsing the non-randomized controlled study method,240 patients with H-type hypertension were divided into test group and control group according to the patient's willingness. Both groups were given Enalapril maleate folic acid tablets, while the test group had modern five elements health exercise, and the control group had walking exercise. Both groups were observed for 12 weeks, and blood pressure, traditional Chinese medicine (TCM) symptom score, quality of life score (Duchenne scale) and homocyteine (Hcy)were observed and compared before and after the treatment.ResultsThe diastolic blood pressure at 4 weeks, systolic and diastolic blood pressure at 8 weeks and systolic blood pressure at 12 weeks in the test group was significantly lowered than those in the control group (P<0.05) The score of Duchenne hypertension quality of life scale at 12 weeks in the test group was more significantly increased than that in the control group (P<0.05). There were no serious adverse events in both groups.ConclusionModern five elements health exercise as the adjuvant treatment has a good effect on H-type hypertension, which can reduce blood pressure and improve the quality of life of patients with H-type hypertension.

      摘要:ObjectiveTo explore the clinical effect of modified Taohong Siwu Decoction and Wuling Powder combined with hydrochlorothiazide on chronic renal failure(CRF)with blood stasis and water damp syndrome.Methods60 CRF patients with blood stasis and water dampness syndrome received by Department of Nephrology of Beijing Hospital of Traditional Chinese Medicine Affiliated to the Capital Medical University and Beijing Pinggu District Hospital of Traditional Chinese Medicine from April 2018 to May 2020 were selected and divided into observation group (n=30) and control group (n=30) . All patients were given the same routine treatment plus orally taking hydrochlorothiazide,and on the basis of treatment in observation group, the control group also combined modified Taohong Siwu Tang plus Wuling Powder treatment, with the treatment course of 2 months. Before and after treatment,TCM syndrome score, 36 Health Survey Synopsis (SF-36), acute physiological and chronic health condition score II (Apache II) , renal function index including serum creatinine (CR) , urea nitrogen (Bun) level and 24h-UTP , inflammatory index including serum interleukin-6(IL-6),tumor necrosis factor factor (TNF-α), c-reactive protein (CRP) concentration were measured.ResultsThe total effective rate of the observation group was 90.00% (27/30) , which was significantly higher than that of the control group (60.00%,18/30)(P<0.01) . Compared with that before treatment, TCM syndrome scores of both two groups were decreased after treatment (P<0.01), and the observation group was lower than the control group (P<0.01). The levels of CR, Bun, IL-6, TNF-α, CRP and 24h-UTP in the two groups after treatment were significantly lower than those before treatment (P<0.05) , but the levels of above index in the observation group were significantly lower than those in the control Group (P<0.01). Compared with that of the control group,SF-36 scores were significantly increased (P<0.01) and Apache II scores were significantly decreased (P<0.01) in both groups after treatment, and SF-36 scores were significantly higher (P<0.01) and Apache II scores were significantly lower (P<0.01) in the observation group than in the control group. The side effects were few and mild in both groups.ConclusionModified Taohong Siwu Decoction and Wuling Powder combined with hydrochlorothiazide can effectively and safely decrease the expression of serum inflammatory factors in CRF patients with blood stasis and dampness syndrome,and improve their renal function.

      摘要:ObjectiveTo explore the features of endowment and disease onset about five circuit phases and six atmospheric influences of patients in Beijing with cerebral infarction.MethodsIt was a retrospective study based on 1327 cases of patients with cerebral infarction who visited the Department of Acupuncture and Moxibustion of Huguosi TCM Hospital of Beijing University of CM from 2015 to 2019. Statistical analysis was performed on the characteristics of the five circuit phases and six atmospheric influences at the time of birth and disease onset of the patient.ResultsIn Beijing,the number of patients born in Taishui year evolutive phase, Jin host evolutive phase,Tu passenger evolutive phase,Yangming Sitian-Shaoyin Zaiquan, Yangming host climatic qi accounts for the largest.During the five years of the included studies,the number of patients with the highest incidence was in Bing Shen year,and the lowest incidence in Yi Wei year.In Yi Wei year, the number of people got cerebral infraction in Jin host evolutive phase,Yangming and Taiyang host climatic qi accounted for the largest,the number of people got cerebral infraction in Mu host evolutive phase,Jueyin and Shaoyin host climatic qi accounted for the least. In Bing Shen year, the number of people got cerebral infraction in Huo host evolutive phase,Jueyin and Shaoyin host climatic qi accounts for the largest,the number of people got cerebral infraction in Jin host evolutive phase,Taiyin host climatic qi accounts for the least.In Ding You year,the number of people got cerebral infraction in Tu host evolutive phase,Shaoyin host climatic qi accounted for the largest, the number of people got cerebral infraction in Shui host evolutive phase,Yangming host climatic qi accounted for the least. In Wu Xu year, the number of people got cerebral infraction in Huo host evolutive phase,Shaoyin host climatic qi accounted for the largest,the number of people got cerebral infraction in Shui host evolutive phase, Yangming host climatic qi accounted for the least. In Ji Hai year, the number of people got cerebral infraction in Huo and Jin host evolutive phase,Taiyin host climatic qi accounted for the largest,the number of people got cerebral infraction in Shui host evolutive phase, Yangming host climatic qi accounted for the least.ConclusionIn Beijing,people born with insufficiency of the heart-yang and with exuberant of liver qi are more likely to suffer from cerebral infarction than other people. The year of Taishu is the year of high incidence of cerebral infarction.The "wood" and "fire" attributes of the five elements of five circuit phases and six atmospheric influences may be one of the external causes of cerebral infarction.
