摘要:ObjectiveTo observe the effect of Wenshen Jianpi Tiaoshu Recipe on the expression of pCREB, Arc and c-fos in colon tissue of diarrhea-predominant irritable bowel syndrome(IBS-D)rats.MethodsAfter adaptive feeding for 5 days,40 Wistar rats were divided into 5 groups with random number table,which were normal group(NG),model group(MG),Chinese medicine treatment groups of high dosage, medium dosage and low dosage(ZHG,ZMG,ZLG),with 8 rats in each group. IBS-D rat models were established by the method of avoiding water stress experiment and senna leaf garage in MG,ZHG,ZMG and ZLG. After modeling,the Chinese medicine treatment groups(ZHG,ZMG,ZLG)were given with high, medium and low doses of Wenshen Jianpi Tiaoshu Recipe by gavage respectively,MG and NG were given normal saline respectively by gavage, all once a day and for 10 days. The general conditions,body mass changes,number of defecation pellets,visceral sensitivity and histopathology were observed. Immunohistochemistry was used to detect the expression of pCREB,Arc and c-fos in colon tissue.ResultsAfter treatment,the body mass of ZHG and ZMG was significantly higher than that of MG(P<0.05);the number of defecation pellets within 1 hour of ZHG,ZMG and ZLG was significantly less than that of MG(P<0.05);the abdominal wall withdrawal reflex(AWR)scores of ZHG was lower than that of MG(P<0.05);Histopathological observation showed mild edema in MG and ZLG mucosa;The expression of pCREB in colon tissue of ZHG,ZMG and ZLG was lower than that of MG(P<0.05);the expression of Arc and c-fos in colon tissue of ZHG and ZMG was lower than that of MG(P<0.05).ConclusionWenshen Jianpi Tiaoshu Recipe can increase the body weight,reduce the number of defecation granules and lower visceral sensitivity of IBS-D rats. Its therapeutic mechanism may be related with the regulation of the expression of pCREB,Arc and c-fos in colon tissue.

摘要:ObjectiveTo evaluate the effectiveness and safety of Shengjiang Runchang Formula on treating functional constipation of spleen deficiency and intestinal dryness type in children.MethodsThe sick children were divided into treatment group and control group by complete randomized control method. The treatment group was given Shengjiang Runchang Formula,and the control group was given Lactulose oral solution. The treatment course of both groups was 3 weeks. And then,the defecation interval time,stool traits,and total effective rate of the two groups were evaluated after the treatment;the relief of poor appetite,abdominal distension,and recurrence rate of the two groups was also evaluated.ResultsAfter treatment,the defecation interval time and stool traits were showed significant improvement(P<0.05);the treatment group was better than the control group(P<0.05),and the total effective rate and the relief of anorexia of treatment group were better than that of the control group and the recurrence rate was lower (P<0.05).ConclusionShengjiang Runchang Formula is safe and effective in treating functional constipation of spleen deficiency and intestinal dryness type in children with low recurrence rate.

摘要:ObjectiveTo observe the effect of added Qishen Taohong Granules on treatment of patients with chronic heart failure (CHF).MethodsUsing prospective,randomized,controlled clinical trial methods,103 CHF patients were randomly divided into treatment group and control group. Both groups were treated with conventional western medicine. On this basis,the treatment group was treated with Qishen Taohong Granules and the control group was treated with trimetazidine hydrochloride for 4 weeks. The changes in cardiac function (NYHA classification),diuretic discontinuation rate and rehospitalization rate of the two groups of patients before and after treatment were evaluated,and the safety of Qishen Taohong Granules in the treatment of chronic heart failure was evaluated.ResultsAfter 4 weeks of treatment,the total effective rate of NYHA cardiac function classification in the treatment group was 76.2%,and that of the control group was 58.8%,and the difference of two groups were statistically significant(P<0.05);the diuretic discontinuation rate of patients in the treatment group was 80.00% and the control group was 52.94 %,there was a statistical difference between the two groups (P<0.05). After 24 weeks of enrollment,the rehospitalization rate of patients in the treatment group and the control group were 33.33% and 46.34% respectively. There were no obvious adverse events in the two groups during the treatment.ConclusionQishen Taohong Granules can improve the cardiac function,increase the diuretic discontinuation rate and reduce the rehospitalization rate of patients with chronic heart failure.
摘要:ObjectiveTo investigate the effect of self-designed formula Longmu Decoction on the methylation level of high-affinity IgE receptor (FcERI) γ subunit promoter and Th2 immune response in mice with atopic dermatitis (AD).MethodsAnimal grouping and modeling:30 BALB/C mice were divided into model group,traditional Chinese medicine group and blank group,with 10 mice in each group. Among them,20 mice in model group and traditional Chinese medicine group were used to make AD model. Traditional Chinese medicine group was given Longmu Decoction by intragastric administration,and the model group and the blank group were given 0.9% normal saline by intragastric administration. All mice were given intragastric administration once a day for 3 weeks. First,the mice in each group were routinely sacrificed by cervical dislocation,femur and tibia of them were obtained,and bone marrow dendritic cells (BMDC)were isolated and cultured;then,the blood of Chinese medicine group and model group mice were immediately collected through the abdominal aorta,the serum was separated,and 10% serum containing Longmu decoction and 10% non-medicated serum were prepared respectively. BMDC was randomly divided into the control group and medicine group. The control group was added with 10% non-medicated serum and the medicine group was added with 10% serum containing Longmu Decoction both continuously cultured for 48 hours.CD11c positive rate of BMDC was detected by flow cytometry. The expression of FCER1γ protein in BMDC cells was detected by Western blotting. Methylation specific PCR (MSP) detection; the levels of McP-1 and IL-12 in the supernatant of BMDC cells were determined by ELISA. The double positive rate of CD4 and IL-4 in BMDC was detected by flow cytometry.ResultsCompared with the control group,the expression of FCER1γ protein in BMDC was lower in Chinese medicine group(P<0.05),and the methylation level of FCER1γ subunit promoter in the drug-containing serum group was higher than that in the control group(P<0.05). Compared with control group,the level of MPC-1 was lower and the level of IL-12 was higher in Chines medicine group(P<0.05). Compared with blank group,there was no significant difference in CD4 and IL-4 double positive rates in Chines medicine group(P>0.05).ConclusionLongmu Decoction can increase the methylation level of FcER Iγsubunit gene regulatory sequence in dendritic cells of AD mice,and further inhibit the overexpression of FcERIγ subunit and FcERI on cell surface,promote Th1 differentiation and inhibit Th2 immune response.
摘要:ObjectiveTo investigate the effect of anxiety on inflammatory factors(NF-κB,TNF-,IL-1)after myocardial infarction in rats and the intervention effect of Shuangxin Prescription.MethodsSD rats were divided into control group,model group,Chinese medicine group and western medicine group according to random number table,with 10 rats in each group. The composite model of anxiety combined with myocardial infarction was established by empty bottle stimulation and ligation of anterior descending branch,and intervention was given by intragastric administration of Shuangxin Prescription. Cardiac function was evaluated by echocardiography,activity ability by field test,anxiety state by elevated cross maze,and levels of NF-κB,TNF- and IL-1 in peripheral blood were detected.ResultsCompared with the control group,the left ventricular ejection fraction of the model group,the Chinese medicine group and the Western medicine group was lower than that of the myocardial infarction group,and the left ventricular ejection fraction of the Chinese medicine group was higher than that of the model group and the western medicine group,with statistical significance in difference(P<0.05). Compared with the control group,the expressions of NF-κB,TNF- and IL-1 in serum of model group,Chinese medicine group and western medicine group were increased,but the expressions of NF-κB,TNF- and IL-1 in serum of Chinese medicine group were lower than those in model group and western medicine group,the differences were statistically significant (P<0.05). Compared with the model group,the anxiety behavior was improved in both the Chinese medicine group and the western medicine group. Conclusion Anxiety can affect the level of inflammatory factors of rats with myocardial infarction and Shuangxin Prescription can reduce the expression of TNF-,IL-6 and IL-1 in serum and promote the cardiac function and relieve anxiety behavior.
