      摘要:ObjectiveTo explore the pharmacological mechanism of Rhizoma coptidis(Rc)in improving diarrhea in ulcerative colitis based on the regulatory effect of aquaporins(AQPs)in colonic mucosal epithelial cells(cmEC).MethodsSD rats were randomly divided into 5 groups,including control group,model group,low,medium and high dose group of Rhizoma coptidis, with 6 rats in each group.Dextran sulfate sodium salt(DSS)was used to establish UC rat model. On the second day after modeling,each group was given by garage the corresponding drugs once a day,while the control and model groups were given the same dose of distilled water. The loose stool rate, the average ranking of loose stool,and the diarrhea index were compared according to the fecal traits and stool times after 7 days of treatment. Hematoxylin-eosin(HE)staining was used to observe the pathological morphological changes of colon tissue; FITC-Dextran was used to detect the colonic mucosal permeability;ELISA kit was used to detect the serum interleukin⁃1β(interleukin⁃1β)and tumor necrosis factor⁃ɑ(TNFɑ)of the rats;the fluorescence quantitative PCR and Western blotting were used to detect the expressions of AQP3 and AQP4 in cmEC of rats.ResultsThe loose stool rate,the average ranking of loose stool,and diarrhea index were significantly increased in the model group(P<0.01),and the colonic mucosal permeability and colonic mucosal inflammatory injury were also considerably increased;meanwhile,the gene and protein expression of AQP3 and AQP4 in cmEC were significantly increased(P<0.01).Compared with the model group,the loose stool rate,average ranking of loose stool,and diarrhea index of the rats in the medium and high dose groups of Rc were significantly ameliorated(P<0.01)with the colonic mucosal permeability and colonic mucosal inflammatory injury improved.At the same time,the gene and protein expression of AQP3 and AQP4 in cmEC were significantly increased(P<0.01).Moreover,the effect of Rc was more significant in the middle-dose group than that in the high dose group of Rc.ConclusionRhizoma coptidis has a certain effect on anti-diarrhea in colitis rats,and its mechanism may be related to reducing the inflammatory injury of UC colonic mucosa,and up-regulating the gene and protein expression of AQP3 and AQP4 in cmEC.

      摘要:ObjectiveTo observe the effect of Qingre Huashi Quyu Recipe on intestinal flora in patients with mild to moderate ulcerative colitis(UC).Methods13 patients with UC in the active stage who were admitted to the Beijing Traditional Chinese Medicine Hospital Affiliated to the Capital Medical University from June 2018 to August 2021 were selected as the research objects. The patients were treated with Qingre Huashi Quyu Recipe for 12 weeks on the basis of long-term mesalazine treatment. The distribution of intestinal flora before and after treatment was detected by 16s rRNA sequencing technology, and the regulatory action of Qingre Huashi Quyu Recipe on intestinal flora was observed.ResultsThe diversity and abundance of flora after the treatment were higher than before;the abundance of Firmicutes and Faecalibacterium was increased, while the abundance of Bacteroides and Escherichia coli decreased.ConclusionQingre Huashi Quyu Recipe can help to increase the diversity and species richness of intestinal flora,assist the growth of beneficial bacteria,and inhibit the proliferation of pathogenic bacteria,so as to make a better maintainence of the intestinal microenvironment.

      摘要:ObjectiveTo explore the therapeutic effect and mechanism of Bushen Zhuyun Decoction(BSZY-D)in the treatment of model rats with luteal phase defect(LPD).MethodsA total of thirty female SD rats were randomly divided into control group,model group,BSZY-D(0.9 g/100 g)group,BSZY-D(1.8 g/100 g)group and dydrogesterone group.After the rats entered the estrous period,all groups were given mifepristone in the morning except the control group,and normal saline in the control group,corresponding drugs in the afternoon,and normal saline in the control and model groups.The rats were killed after LPD modeling and administration for 3 consecutive estrous cycles.The levels of serum estradiol(E2)and progesterone(P),and the mRNA as well as the protein levels of the three neuropeptides and their receptors(Kisspeptin/GPR54,NKB/NK3R and DYN/KOR)were detected.ResultsCompared with the control group,the levels of serum E2 and P in the model group were decreased,but the difference was not statistically significant(P>0.05);compared with the model group,the levels of E2 and P in BSZY-D(0.9g/100g)group and BSZY-D(1.8 g/100 g)group were increased,and there was significant difference in BSZY-D(0.9 g/100 g)group,BSZY-D(1.8 g/100 g)group with E2(P<0.05).The mRNA levels of KNDy neurons related peptide in the model group were decreased compared with the control group,the difference of GPR54,NK3R and DYN were statistically significant(P<0.05);compared with the model group,the related genes in BSZY-D(0.9 g/100 g)group and BSZY-D(1.8 g/100 g)group showed an upward trend,and there was significant difference in GPR54,NK3R,and DYN in BSZY-D(0.9 g/100 g)group and BSZY-D(1.8 g/100 g)group(P<0.05).The protein expression of KNDy neurons related peptide in model group was decreased compared with control group,and the differences of NKB,DYN and KOR were statistically significant(P<0.05);compared with the model group,the KNDy neuron related protein in BSZY-D(0.9 g/100 g)group and BSZY-D(1.8 g/100 g)group showed an upward trend,and there was significant difference in GPR54,NKB,NK3R,DYN and KOR in BSZY-D(1.8 g/100 g)group(P<0.05).ConclusionBushen Zhuyun Decoction might improve ovarian luteal endocrine by up regulating the expression of KNDy neurons related neuropeptide in rats,so as to have a therapeutic effect on LPD.

      摘要:ObjectiveTo study the effect of Jianpi Liqi Decoction on the improvement of micro-inflammation in duodenum of rats with functional dyspepsia(FD).Methods24 SD male rats were randomly divided into normal group(6 rats)and model group(18 rats).The FD rat model was established by tail-clamping stress.Then the model group was randomly divided into FD model group,Chinese herbal medicine treatment group and CRH antagonist group.The normal and model group were given pure water by intragastric administration,while the Chinese herbal medicine treatment group was given Jianpi Liqi Decoction by intragastric administration,for 7 days and the CRH antagonist was injected intraperitoneally to the antagonist group before tail-clamping stress was used at 8 o'clock every day from the 1st day of modeling till the end of modeling.After the intervention finished,the deudenum,hypothalamus,and serum of the rats in each group were taken,and HE staining was used to observe the basic structure of duedenum,and Western blotting and ELISA to measure the content of Corticotropin-Releasing Hormone(CRH)in the hypothalamus,duedenum and serum,and Western blotting to measure the content of substance P(SP),eosinophilic major basic protein(MBP),and the expression of tryptase protein in mast cells in the duodenal tissues.ResultsThere was no obvious abnormality in the structure and morphology of duodenum,ulcer,intestinal villi destruction and obvious inflammation in each group were not seen.There was no significant difference in the CRH content in the hypothalamus of rats in each group(P>0.05).The content of CRH in duodenum and serum of model group was higher than that of normal group(P<0.05).The content of CRH in duodenum and serum in the treatment group was lower than that in the model group(P<0.05).Serum CRH content in antagonist group was lower than that in model group (P<0.05),but there was no significant difference in duodenal CRH content between antagonist group and model group(P>0.05).SP content,MBP and Tryptase protein expression in duodenum of model group were higher than those of normal group(P<0.05).The SP content,MBP and Tryptase protein expression in duodenum of the treatment group were lower than those of the model group(P<0.05).The expression of Tryptase protein in duodenum of antagonist group was lower than that of model group(P<0.05),and SP content and MBP protein expression were lower than those of model group,but the difference was not statistically significant(P>0.05).ConclusionJianpi Liqi Decoction may improve local duodenal micro-inflammation in duodenum,such as eosinophils and mast cell infiltration,and have a therapeutic effect on FD by inhibiting SP-mediated activation of the brain gut axis under stress.

      摘要:ObjectiveAn UPLC-MS/MS method was established to study the effects of different origin, harvest period, tree age and processing methods on caffeine content in Ginkgo Folium.MethodsThe sample was extracted by ultrasound with methanol,steamed drying solvent,alkalized after dissolution of acid water and chloroform extraction 3 times,and separated by ACQUITY UPLC HSS T3 chromatographic column,with mobile phase of 0.1% formic acid solution-0.1% formic acid acetonitrile solution for gradient elution.Data were conducted at ESI+ multiple reaction monitoring(MRM)mode;quantitative analysis was conducted with external standard method.And the effects of origin,harvesting period,tree age and processing method on caffeine content in Ginkgo Folium was studied by statistics.ResultsWith caffeine in the range of 0.06 ng/mL~ 62.5 ng/mL,there was a good linear relationship between the mass concentration of caffeine and the peak area,R2=0.9999;the sample recovery rate was 84.94%,with RSD of 7.99%.The content range of caffeine in 77 batches of Ginkgo Folium sample was 0.1585~22.1894 μg/kg.The caffeine content of Ginkgo Folium produced in Ningqiang,Shaanxi province and Changting,Fujian province,was highest,second in Pizhou,Jiangsu Province,Tancheng,Shandong Province and Dandong,Liaoning Province,and lowest in Wenshan,Yunnan Province.ConclusionThe content of caffeine in most Ginkgo Folium is stable at a lower level(below 10 μg/kg).The content of caffeine in Ginkgo Folium is not significantly related with the tree age,harvesting period and processing method,but with the origin of Ginkgo Folium.

      摘要:Thrombocytopenia is a common complication of chronic liver disease, the patients can have the coagulation mechanism damaged,causing bleeding in the skin, mucous membranes and even organs,which may affect the prognosis and become life-threatening in severe cases. Ying (nutrient blood) and Wei (defensive qi) are originated from water and food. Ying takes Qi as the nature, and Wei takes blood as the nature. When Ying and Wei is in harmony, the body will be in a constant state of balance. The pathogenesis of thrombocytopenia in liver cirrhosis is caused by the disharmony between Yin and Wei. On the one hand,the lack of harmony between Yin and Wei is due to stagnation of Wei qi and hesitant flow of Ying blood , making blood difficult to flow along the meridians and then out of the vessles. Secondly, deficiency of Qi in the middle jiao and insufficient production of qi and blood may lead to the defect of blood quality. The syndrome differentiation and treatment should focus on grasping the key points of pathogenesis, clarifying the impassability or insufficiency, and harmonizing the middle jiao as the basic therapeutic principle.Clinically, Guizhi Decoction and other decoctions in same category can be used and modified according to the changes of the disease in order to regulate Ying Qi and Wei blood, tonify the middle jiao, and the clinical and experimental evidence should also be considered to ensure the efficacy.It has guiding significance for the treatment of liver cirrhosis and thrombocytopenia.

      摘要:ObjectiveTo observe the clinical effect of Jieyu Prescription combined with in the treatment of post-stroke depression and neural functional recovery for Ischemic Stroke.Methods76 patients with post-stroke depression were randomly divided into treatment group(n=38)and control group(n=38).The treatment group was treated with Jieyu formula plus acupuncture on the basis of conventional treatment,while the control group was treated with fluoxetine Capsule on the basis of conven-tional treatment for 4 weeks.The changes of HAMD⁃17 scale,SDS scale,NIHSS score,ADL scale,serum IL⁃1 and IL⁃6 were detected before and after treatment.ResultsAfter treatment,HAMD⁃17 scale,SDS scale and NIHSS scores of the two groups were lower than before treatment,the treatment group was significantly better than the control group(P<0.05),with moderate effect(0.5<Cohen's D effect<0.8),SDS scale with higher effect(Cohen's D effect=0.8);ADL scale of the two groups were higher than before treatment,the treatment group was significantly better than the control group(P<0.05),with moderate effect(0.5<Cohen's D effect<0.8).The total effective rate of the treatment group was 89.74%,which was better than 76.32% of the control group(P<0.05);the plasma concentrations of IL⁃1 and IL⁃6 in the two groups were lower than before treatment,and the treatment group was significantly better than the control group(P<0.05),with moderate effect(0.5<Cohen's D effect<0.8).ConclusionJieyu formula combined with acupuncture can promote the recovery of neurological function in patients with ischemic stroke and improve post-stroke depression.Its mechanism may be related to the regulation of plasma IL⁃1 and IL⁃6 concentrations.

      摘要:ObjectiveTo investigate the effect of Yishen Zhongzi Presription on asthenospermia patients with kidney and essence deficiency.Methods140 patients with asthenospermia were randomly divided into two groups according to the random number table method,70 cases in treatment group and 70 cases in control group.The treatment group was treated with Yishen Zhongzi Prescription,and the control group was treated with Weishen Zinc Capsule.Four weeks was one course of treatment. After three courses,the clinical efficacy,sperm motility,sperm DFI,TCM symptom score and other main parameters of the two groups before and after the treatment were observed.ResultsAfter the treatment, the effective rate was 80.60% in the treatment group and 55.38% in the control group, the difference was statistically significant(P<0.05).The sperm motility of two groups after the treatment was higher than that before the treatment(P<0.05),and the treatment group was better than the control group(P<0.05).After the treatment,the DFI of the treatment group was decreased(P<0.05),which was lower than that of the control group.During the treatment period,2 cases in the experimental group had gastric distension after taking the medicine on an empty stomach,and then changed to take the medicine 1h after meal,and gastric distension disappeared without any other discomfort;in the control group,3 cases had gastric distension after taking the medicine,and the patients stopped taking the medicine by themselves,so they fell off the research.There was no abnormality in blood and urine routine,liver and kidney function in both groups.ConclusionYishen Zhongzi Prescription can improve sperm motility and reduce DFI in patients with asthenospermia with kidney and essence deficiency with good clinical effect and safety.
