Objective To observe the effects of Jianpi Yishen Tongluo Recipe on NF-κB,caspase-3 and inflammatory factors in mice with MPTP-induced Parkinson’s disease( PD). Methods MPTP was intraperitoneally injected into mice to generate PD model. Then they were randomly divided into 5 groups: normal group, model group, Jianpi Yishen Tongluo Recipe group( JPTQ),NF-κB inhibitor group( PDTC),and Jianpi Yishen Tongluo Recipe + NF-κB inhibitor group( JPTQ + PDTC). The Chinese herbal medicine groups were given Jianpi Yishen Tongluo Recipe by intragastric administration for 2 weeks. The behavioral changes were evaluated by pole climbing test. The protein expressions of NF-κB p65 and caspase-3 in brain tissue were detected by western blot. The contents of TNF-α and IL-6 in striatum were detected by ELISA. Results Compared with the normal group,the climbing time of each group was increased significantly on the 7 th day of MPTP injection( P < 0. 01). On the 21 st day,the climbing time of the MPTP model groups were still longer( P < 0. 01). Compared with the model group, protein expression of NF-κB p65 and caspase-3 in brain tissue,the contents of TNF-α and IL-6 of Jianpi Yishen Tongluo Recipe group and Jianpi Yishen Tongluo Recipe + PDTC group were decreased in striatum( P < 0. 01). There was no significant difference between the two groups( P > 0. 05). Conclusion Jianpi Yishen Tongluo Recipe has a protective effect on MPTP-induced neuronal injury in mice with Parkinson’ s disease. Its mechanism may be related to reduction of NF-κB and Caspase-3 protein expression,leading to alleviation of inflammation and decrease of apoptosis.
