目的观察慈姑多糖对高脂饮食诱导的非酒精性脂肪肝病(NAFLD)小鼠糖脂代谢的调节作用,并探讨其可能的作用机制。方法将26只SPF级健康雄性ICR小鼠随机分为3组,对照组(8只)、多糖组(8只)和模型组(10只)。多糖组与模型组采用高脂饲料喂饲建立NAFLD模型,同时多糖组给予慈姑多糖灌胃干预12周。采用全自动生化分析仪检测血清甘油三酯(TG)、总胆固醇(TC)、空腹血糖(FBG)、游离脂肪酸(FFA)水平; HE染色、油红O染色观察肝脏的组织形态学特点;用荧光定量PCR法检测肝组织肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、过氧化物酶体增殖物活化受体γ协同激活因子-1α(PGC-1α) mRNA的表达。结果与对照组相比,模型组血清TG、TC、FBG、FFA水平高(P <0. 01);肝组织呈中度脂肪变性,同时伴有炎症和坏死;肝组织中TNF-α、IL-6 mRNA表达高(P <0. 05),PGC-1αmRNA表达低(P <0. 01)。与模型组相比,多糖组血清TG、TC、FPG、FFA水平低(P <0. 05);肝组织TNF-α、IL-6 mRNA表达低(P <0. 05,P <0. 01),PGC-1αmRNA表达高(P <0. 01)。结论慈姑多糖能够明显改善NAFLD小鼠糖脂代谢,其作用机制可能与上调PGC-1αmRNA表达,降低TNF-α、IL-6 mRNA表达有关。
Objective To observe the effect of Sagitta sagittifolia polysaccharide on glucose and lipid metabolism in mice with nonalcoholic fatty liver disease( NAFLD) induced by high fat diet and discuss its possible action mechanism. Methods SPF grade ICR mice( n = 26) were randomly divided into three groups: control group,model group,and polysaccharide group( n = 8 in control and polysaccharide group,n = 10 in model group). The mice in the model and polysaccharide groups were fed with high fat diet( MCD) to establish NAFLD model,meanwhile,the polysaccharide group was given Sagitta sagittifolia polysaccharide. Serumtriglyceride( TG),total cholesterol( TC) fasting blood-glucose( FBG) and free fatty acids( FFA) were tested by full automatic biochemical analyzer. The histomorphology of liver was observed by HE staining and oil red 0 staining,and the expressions of TNF-α,IL-6,PGC-1α mRNA in liver tissue were detected by PCR. Results Compared with the control group,the serum levels of TG,TC,FBG and FFA in the model group were higher than those in the control group( P < 0. 01). Liver tissue showed moderate steatosis accompanied by inflammation and necrosis,and the expression of TNF-α,IL-6 mRNA was increased in liver tissue( P < 0. 05). The expression of PGC-1α mRNA was decreased( P < 0. 01). Compared with the model group,the level of serum TG,TC,FBG and FFA in polysaccharide group decreased( P < 0. 05). The expression of TNF-α and IL-6 mRNA was decreased in liver tissue( P < 0. 05 or P < 0. 01). The expression of PGC-1α mRNA was increased( P < 0. 01). Conclusion Sagitta sagittifolia polysaccharide can significantly regulate the metabolism of glucose and lipid in NAFLD mice,its mechanism may be related with the up-regulation of the expression of PGC-1α mRNA,and decrease of the expression of TNF-α and IL-6 mRNA.
