目的,2,基于肿瘤炎性微环境NF-κB信号通路及炎性细胞因子,探究肺瘤平膏防治肺癌的效应机制。,方法,2,采用SPF级C57BL/6雄性小鼠进行Lewis肺癌移植瘤造模,分为空白对照组、模型对照组、肺瘤平膏组、环磷酰胺(Cyclophosphamide,CTX)组、肺瘤平膏+CTX组共5组。肺瘤平膏组给予肺瘤平膏灌胃,CTX组给予CTX腹腔注射,肺瘤平膏+CTX组给予以上2种干预措施。各组均干预21 d后处死,称瘤体质量,计算抑瘤率;免疫组织化学法检测肿瘤组织中NF-κB、E-cadherin、N-cadherin表达情况;ELISA法检测小鼠血清中炎性细胞因子IL-6、TNF-α和IL-1β的水平。,结果,2,肺瘤平膏可抑制Lewis肺癌移植瘤肿瘤生长达38.8%,而与CTX联合用药可抑制肿瘤生长达83.2%;与模型对照组比较,肺瘤平膏显著降低血清中促炎细胞因子IL-6、TNF-α和IL-1β的水平(,P,<,0.01),抑制肿瘤组织中NF-κB、N-cadherin的表达,促进E-cadherin的表达(,P,<,0.01)。,结论,2,肺瘤平膏可抑制肿瘤炎性微环境中关键信号NF-κB及炎性细胞因子水平,进而调控上皮间质转化,提示肺瘤平膏可能是炎性相关肺癌防治的有效方药。
Objective,2,To explore the effect mechanism of Feiliuping Ointment on the prevention and treatment of lung cancer based on NF-κB signaling pathways and inflammatory cytokines in tumor inflammatory microenvironment.,Methods,2,SPF grade C57BL/6 male mice were modeled by Lewis lung cancer cell transplanted tumor, and divided into 5 groups, that is blank control group, model control group, Feiliuping Ointment group, cyclophosphamide (CTX) group, and CTX + Feiliuping Ointment group. The mice were given Feiliuping Ointment gavage or intraperitoneal injection of CTX for 21 days respectively, and then sacrificed. The tumor was weighed to calculate the tumor inhibition rate;the expression of NF-κB, E-cadherin and N-cadherin in tumor tissue was detected by immunohistochemistry;the levels of inflammatory cytokines IL-6,TNF-α and IL-1β in the serum of mice were detected by ELISA.,Results,2,Feiliuping Ointment can inhibit the growth of Lewis lung cancer xenograft tumor by 38.8%, while the combination with CTX can inhibit the tumor growth by 83.2%;compared with the model group,Feiliuping Ointment significantly reduced the pro-inflammatory cytokines IL-6,TNF-α and IL-1β in serum (,P,<,0.01);compared with the model group,Feiliuping Ointment inhibited the expression of NF-κB,promoted the expression of E-cadherin and inhibited the expression of N-cadherin in tumor tissue (,P,<,0.01).,Conclusion,2,Feiliuping Ointment can inhibit the NF-κB signaling pathway and the levels of inflammatory cytokines in tumor inflammatory microenvironment,thereby regulating epithelial-mesenchymal transition, suggesting that Feiliuping Ointment may be an effective prescription for the prevention and treatment of inflammation-related lung cancer.
