目的,2,观察菖蒲郁金汤化裁方对慢加急性肝衰竭(acute-on-chronic liver failure,ACLF)大鼠脑损伤所致精神障碍的影响及机制。,方法,2,30只Wistar大鼠随机分为对照组、模型组和中药组,采用人血清白蛋白免疫诱导大鼠肝纤维化,中药组予以菖蒲郁金汤化裁方灌胃5 d,对照组、模型组均予以0.9%氯化钠注射液灌胃5 d,再予D-氨基半乳糖(D-GalN)/脂多糖(LPS)联合腹腔注射急性攻击,建立ACLF大鼠模型。各组均在急性攻击8 h后麻醉处死。HE染色观察各组肝、脑组织病理变化,电镜观察各组脑组织超微结构变化,Western blotting法检测脑组织抗淀粉样前体蛋白(APP)、突触素(SYN)、生长相关蛋白-43(GAP-43)的表达。,结果,2,与模型组相比,中药组ACLF大鼠神经元及突触超微结构的损伤减轻,脑组织内APP表达降低(,P,<,0.05),SYN和GAP-43表达提高(,P,<,0.05)。,结论,2,通过抑制APP聚集,提高内源性突触相关蛋白表达,保护突触功能并提高其可塑性,可能是菖蒲郁金汤化裁方改善大鼠ACLF后脑损伤所致神经功能障碍的作用机制。
