目的,2,观察优化新生脉散方对心力衰竭(简称“心衰”)小鼠心功能、运动耐量的影响及其作用机制。,方法,2,将100只清洁级C57BL小鼠随机分为假手术组(,n,=10)和造模组(,n,=90),造模组采用主动脉弓缩窄(TAC)术建立心衰模型。采用随机数字法将造模成功心衰小鼠(LVEF≤45%)50只分为分为模型(Model)组、优化新生脉散方低剂量(YQHXLS-L)组、优化新生脉散方中剂量(YQHXLS-M)组、优化新生脉散方高剂量(YQHXLS-H)组、氯沙坦(Losartan)组,各10只。YQHXLS-L组、YQHXLS-M组、YQHXLS-H组分别按生药量1.545、3.090、6.180 g/(kg·d)灌胃,Losartan组按6.5 mg/(kg·d)灌胃,假手术组及Model组给予等体积0.5%羧甲基纤维素钠悬浊液灌胃,1次/d,连续给药4周。对比各组左室后壁厚度(LVPW)、左室射血分数(LVEF)、左室短轴缩短率(FS)、力竭游泳时间、心脏体质量比,心肌组织形态学改变和心肌纤维化程度,小鼠心肌、腓肠肌中肌肉萎缩F-box蛋白(Atrogin-1) mRNA及蛋白表达。,结果,2,与Model组比较,YQHXLS-L组、YQHXLS-M组、YQHXLS-H组LVEF、FS高,LVPW低、心脏体质量比及小鼠腓肠肌Atrogin-1蛋白表达低(,P,<,0.05),心肌组织病理损伤及纤维化程度改善。其中与Model组比较,YQHXLS-H组力竭游泳时间长,心肌及腓肠肌Atrogin-1 mRNA表达低(,P,<,0.05);YQHXLS-M组、YQHXLS-H组心肌组织Atrogin-1蛋白表达低(,P,<,0.05)。,结论,2,优化新生脉散方可显著改善心衰小鼠的心功能及运动耐量,抑制心肌纤维化及腓肠肌萎缩,其作用机制可能与下调心肌、腓肠肌细胞Atrogin-1表达有关。
