目的

探究土贝母苷甲(TBMS1)对三阴性乳腺癌(TNBC)细胞系MDA-MB-468的影响,并探求其作用机制。
方法

通过MTT实验测定TBMS1对细胞活性的影响并确定给药浓度,通过EDU细胞增殖实验测定TBMS1对细胞增殖能力的影响,通过细胞划痕实验测定TBMS1对细胞迁徙能力的影响,通过Transwell实验测定TBMS1对细胞侵袭能力的影响,通过Western Blot实验测定TBMS1作用于MDA-MB-468细胞的具体作用机制。
结果

TBMS1对MDA-MB-468细胞的增殖、迁徙和侵袭能力呈剂量依赖性抑制。而Western Blot及挽救实验结果表明:TBMS1通过抑制AKT/NF-κB信号通路发挥作用。
结论

TBMS1作为一种天然药物成分,在TNBC的治疗中显示出潜在的抗肿瘤活性,且具有较低的药物毒性。
