研究补肺活血胶囊对慢性阻塞性肺疾病(简称“慢阻肺”)大鼠模型气道炎症及黏液高分泌的干预作用和机制。
方法

采用烟熏并气道内滴注脂多糖的方法制备慢阻肺大鼠模型。40只Wistar大鼠随机分为空白对照组、模型组、补肺活血组和茶碱组,空白对照组及模型组予以生理盐水灌胃,补肺活血组和茶碱组分别予补肺活血胶囊和茶碱缓释片溶液灌胃。使用小动物肺功能仪检测大鼠肺功能,苏木精-伊红染色和阿利新蓝-过碘雪芙染色进行形态学分析,酶联免疫吸附实验检测肺泡灌洗液中IL-1β、IL-6、NE、TGF-β、EGF、IL-13和TNF-α的表达,免疫组化法检测大鼠肺组织中黏蛋白5AC(Muc5ac)的表达,免疫印迹法检测Toll样受体4(TLR4)、髓样分化初级应答基因88(MyD88)、NF-κB、Muc5ac、EGFR/p-EGFR、PI3K/p-PI3K、AKT/p-AKT的蛋白表达。

Objective

To investigate the intervention effect and mechanism of Bufei Huoxue Capsule on airway inflammation and mucus hypersecretion in rats with chronic obstructive pulmonary disease (COPD) .
Methods

The rat model of COPD was established by smoking and intratracheal instillation of lipopolysaccharide . The lung function was measured by small animal lung function meter, and the morphology of airway epithelial cells was detected by hematoxylin-eosinstaining and alicinblue-periodopho staining, the expressions of IL-1β, IL-6, NE, TGF-β, EGF, IL-13 and TNF-α in BALF were detected by enzyme-linked immunosorbent assay , the protein expressions of TLR4, Myd88, NF-κB, Muc5ac, EGFR/p-EGFR, PI3K/p-PI3K, Akt/p-AKT were detected by Western-blot, and the expression of Muc5ac was detected by immunohistochemistry.
