目的

基于晚期糖基化终产物(AGEs)/碱性成纤维细胞生长因子(bFGF)信号通路,探讨复方黄柏液促进糖尿病足溃疡(DFU)创面愈合的作用机制。
方法

采用随机数字表法将SPF级雄性大鼠分为空白组、模型组、生长因子组、复方黄柏液组、生长因子+复方黄柏液组(联合组)。除空白组外,均采用高糖高脂饲养,并联合链脲佐菌素(STZ)腹腔注射制备糖尿病模型、DFU模型,给予相应药物治疗,最终取材80只。在DFU造模后第7、14天比较各组创面愈合率,苏木精-伊红(HE)染色、Masson染色观察大鼠创面组织病理学变化,免疫荧光检测创面组织bFGF、成纤维细胞生长因子受体(FGFR)、增殖细胞核抗原(PCNA)表达,酶联免疫吸附试验(ELISA)检测创面皮肤组织血管内皮生长因子(VEGF)、bFGF含量及AGEs-晚期糖基化终末产物受体(RAGE)信号通路相关因子。
结果

第7、14天,与空白组比较,模型组创面愈合率低(

0.05)。与模型组比较,复方黄柏液组、生长因子组、联合组创面愈合率高(

0.05);病理结果显示,各药物组均能促进新生表皮形成和胶原纤维的沉积,减少炎症细胞浸润,加速溃疡的愈合。
结论

复方黄柏液可通过抑制AGEs-bFGF交联通路,影响创面中VEGF、bFGF、PCNA、FGFR表达,促进DFU大鼠创面血管新生,加速创面修复和愈合。
Objective

To investigate the mechanism by which
Compound Huangbai Liquid
promotes the healing of diabetic foot ulcer (DFU) wounds in rats based on the advanced glycation end products (AGEs)/basic fibroblast growth factor (bFGF) signaling pathway.
Methods

SPF-grade male rats were randomly divided using a random number table into five groups: blank group, model group, growth factor group,

group, and growth factor +
Compound Huangbai Liquid
group. Except for the blank group, all rats were fed a high-sugar, high-fat diet and received an intraperitoneal injection of streptozotocin (STZ) to establish diabetes and DFU models. The corresponding treatments were administered, and tissue samples were collected from 80 rats. Wound healing rates were compared on days 7 and 14 after DFU modeling. Histopathological changes in wound tissues were observed using hematoxylin-eosin (HE) and Masson staining. Immunofluorescence was used to detect the expression of bFGF, fibroblast growth factor receptor (FGFR), and proliferating cell nuclear antigen (PCNA) in wound tissues. Enzyme-linked immunosorbent assay (ELISA) was employed to measure the levels of vascular endothelial growth factor (VEGF), bFGF, and factors related to the AGEs-RAGE signaling pathway in wound skin tissue.

On days 7 and 14, compared with the blank group, the model group showed significantly lower wound healing rates (

0.05). Compared with the model group, the
Compound Huangbai Liquid
group, growth factor group, and combination group all exhibited significantly higher wound healing rates (
P
<
0.05). ELISA results showed that levels of AGEs and RAGE in wound skin tissue were significantly decreased (
P
<
0.05), while VEGF and bFGF levels were significantly increased (
P
<
0.05). Immunofluorescence revealed significantly increased expression of bFGF, FGFR, and PCNA in all treatment groups (
P
<
0.05). Histological analysis showed that all treatment groups promoted new epidermis formation and collagen fiber deposition, reduced inflammatory cell infiltration, and accelerated ulcer healing.
Conclusion

Compound Huangbai Liquid
can promote angiogenesis and accelerate wound repair and healing in DFU rats by inhibiting the AGEs-bFGF crosslinking pathway and regulating the expression of VEGF, bFGF, PCNA, and FGFR in the wound.
