摘要

目的

用番泻叶灌胃联合三硝基苯磺酸（TNBS）/乙醇混合物灌肠构建脾肾阳虚型溃疡性结肠炎（UC）大鼠模型，并从以方测证、方证不符2方面验证模型。

方法

第一阶段：番泻叶灌胃6周+TNBS/乙醇混合物灌肠后评价空白（A）组和造模大鼠的证候指标和疾病指标变化情况；第二阶段：将造模大鼠随机分为模型（B）组、以方测证（C）组、方证不符（D）组，各10只，分别灌胃饮用水、温肾健脾方、清热化湿调枢方后，进一步验证证候指标［进食量、体质量、握力、负重游泳时间、肛温，血清D-木糖、促肾上腺皮质激素（ACTH）、皮质酮/肾上腺酮（CORT）水平］和疾病指标［疾病活动指数（DAI）、组织学评分、结肠长度］变化情况。

结果

第一阶段：与A组比较，造模大鼠进食量、体质量、握力、负重游泳时间、肛温降低（

0.05），血清D-木糖、ACTH、CORT水平降低（

0.05），疾病指标（DAI、组织学评分）增高（

0.05），结肠长度缩短（

0.05）。第二阶段：与A组比较，B、C、D组大鼠进食量、体质量、握力、负重游泳时间仍降低（

0.05），血清中D-木糖、ACTH、CORT水平仍降低（

0.05）。C组各项证候指标（除肛温）和疾病指标均较B组改善，差异有统计学意义（

0.05）；D组各项指标虽较B组改善，但差异无统计学意义（

0.05）。

结论

高剂量番泻叶灌胃联合TNBS/乙醇混合物灌肠可成功建立脾肾阳虚证UC大鼠模型，并且具有造模步骤相对简单、安全性高的优点。

Abstract

Objective

To establish a rat model of ulcerative colitis（UC）with spleen and kidney

yang

deficiency syndrome by intragastric administration of high-dose Senna leaves combined with trinitrobenzene sulfonic acid（TNBS）/ethanol enema，and to validate the model from the perspectives of "prescription testing the syndrome" and "prescription-syndrome discrepancy".

Methods

In the first stage，changes in syndrome and disease indices were evaluated in a blank control group（A）and model rats after 6 weeks of Senna leaf gavage combined with TNBS/ethanol enema.In the second stage，the model rats were randomly divided into a model group （B），a prescription-testing-syndrome group（C），and a prescription-syndrome discrepancy group（D），each with 10 rats.Rats in groups B，C，and D received drinking water，

Wenshen Jianpi Formula

，and

Qingre Huashi Tiaoshu Formula

，respectively，by gavage，and changes in syndrome and

disease indices were further evaluated.

Results

In the first stage， compared with group A，model rats showed reduced food intake，body weight，grip strength，weight-bearing swimming time，and anal temperature （

0.05）；serum D-xylose，ACTH，and CORT levels decreased（

0.05）；disease indices（DAI，histological score）increased （

0.05）；and colon length was shortened（

0.05）.In the second stage，compared with group A，rats in groups B，C，and D still had decreased food intake，body weight，grip strength，and weight-bearing swimming time（

0.05），as well as lower serum D-xylose，ACTH，and CORT levels（

0.05）.Compared with group B，all syndrome indices（except anal temperature）and disease indices in group C were significantly improved（

0.05），whereas indices in group D improved but without statistical significance（

0.05）.

Conclusion

Intragastric administration of high-dose Senna leaves combined with TNBS/ethanol enema can successfully establish a UC rat model with spleen and kidney

yang

deficiency syndrome.The modeling procedure is relatively simple to implement and demonstrates high safety.
