目的,2,观察参附黄方对脓毒症小鼠生存率、组织损伤及炎症反应的影响。,方法,2,以C57BL/6J雄性小鼠为实验对象,通过盲肠结扎穿孔术(CLP)复制小鼠脓毒症模型。将205只小鼠分为假手术组15只、造模组190只,造模后24 h小鼠死亡64只,存活126只。从造模后存活的小鼠中取54只,随机分为模型组、参附黄高剂量组、参附黄低剂量组各18只,用于观察术后7 d内的生存率;将剩余的72只小鼠随机分为模型组、参附黄高剂量组各36只,造模后72 h将小鼠麻醉后处死取材,观察肠道组织病理变化,并对肺、肠、心、肝、脾、肾组织损伤情况进行评分,ELISA法测定血清TNF-α、IL-1β。各组小鼠造模24 h后给药,模型组给予0.5 mL生理盐水灌胃,参附黄低、高剂量组分别给予0.1、0.5 g/mL参附黄水煎剂0.5 mL灌胃,均每日1次,持续3 d。,结果,2,模型组与参附黄低剂量组各时间点小鼠生存率比较差异无统计学意义(,P,>,0.05);术后3、7 d,参附黄高剂量组小鼠生存率高于模型组(,P,<,0.05)。与假手术组相比,模型组小鼠肺、肠、心、肝、脾、肾组织损伤严重,血清TNF-α、IL-1β水平高(,P,<,0.05)。与模型组相比,参附黄高剂量组肺、肠、心、肝、脾、肾组织损伤程度明显减轻,血清TNF-α、IL-1β水平低(,P,<,0.05)。,结论,2,参附黄方可以通过减轻肺、肠、心、肝、脾、肾组织损伤,抑制炎症因子过度激活,提高脓毒症小鼠生存率。
Objective,2,To observe the effect of Shenfuhuang Formula on the survival rate,tissue damage and inflammatory reaction in septic mice.,Methods,2,The sepsis model of C57BL/6J male mice was established by cecal ligation and puncture(CLP).A total of 205 mice were divided into sham operation group of 15 and model-establishing group of 190. 24 hours after modelling,64 mice were dead and 126 were survived. 54 survived mice were selected and randomly divided into model group,Shenfuhuang high dosage group and Shenfuhuang low dosage group of 18 each. The survival rate of mice in 7 days after CLP was recorded. The rest of 72 mice were randomly divided into model group and Shenfuhuang high dosage group of 36 each. 72 hours after modeling,the mice were put to death after anesthesia to observe the pathological change of intestine and the injury of lung,intestine,heart,liver,spleen and kidney and scored. The expression of serum TNF-α and IL-1β were detected by ELISA. The mice in each group were administrated 24 hours after modeling. The model group was given 0.5 mL saline by gavage,and Shenfuhuang low and high dose groups were given 0.5 mL Shenfuhuang Decoction by gavage of 0.1 and 0.5 g/mL, once a day for 3 days.,Results,2,There was no significant difference in survival rate at different time point between model group and Shenfuhuang low dose group(,P,>,0.05); on the 3rd and 7th day after operation, the survival rate of mice in Shenfuhuang high dose group was higher than that in model group(,P,<,0.05). Compared with the sham group, the lung, intestine, heart, liver, spleen and kidney tissues of the model group were seriously damaged, and the serum TNF-α and IL-1β levels were high(,P,<,0.05). Compared with the model group, the damage degree of lung, intestine, heart, liver, spleen and kidney in Shenfuhuang high dose group was significantly reduced, and the levels of serum TNF-α and IL-1β were low(,P,<,0.05).,Conclusion,2,Shenfuhuang Formula can improve the survival rate of septic mice by reducing the injury of lung, intestine, heart, liver, spleen and kidney, and inhibiting the excessive activation of inflammatory factors.
